| Added different concentrations of WY14643(peroxidase value-added content activated receptor alpha activator) to observe their effects on the in vitro development of early embryos in mouse, and tested the content of hydrogen peroxide (H2O2) in each treatment group, therefore investigated whether WY14643can play an effective antioxidant effect on early embryos in vitro culture. This study can lay the foundation for the antioxidant mechanism of oocytes and early embryos in mammalian. Results are as follows:1ã€In the H2O2minimum lethal concentration selection experiments, the results showed that10-7%of H2O2was the minimum lethal concentration in the process of embryonic development in mouse.2ã€In the comparative experiments of emhryo development rates and H2O2levels between WY14643treatments for2h and4h, at the same concentration,rates of cleavage,4-cell and blastocyst in2h group were all higher than4h group, H2O2, levels in2h group were less than4h group.3ã€In the experiments of different H2O2concentrations combined with WY14643treatments for2h, to detect the rates of embryo development and levels of H2O2,development rates of10-7%+0.1μMã€10-7%+1.0u M M were significantly higher than10-8%H2O2treatment group.Conclusion:All results show that the concentration of0.1u M WY14643,2h can really improve the quality of embryo development,and reduce the generation of reactive oxygen species, improve the efficiency of antioxidant. effectively. |
