Oil pollution of saline-alkli soil has become one of the global environmentproblems. In the present study, we take alfalfa and triticale as materials, analyse the speciesdiversity, functional diversity and genetic diversity of plants in polluted soil at seeding stage,vegetating stage and senescing stage by use of dilution plate, Biolog Ecoplate, PCR-DGGE,determination of oil degradation rate and pot experiment. The main contents are asfollows:1. During the whole growth period of alfalfa and triticale, rhizospheremicroorganisms that can be cultured were less in seeding stage, and began to increasequickly in vegetative stage, whereas it has begun to decrease in senescing stage. Changeof the number of rhizosphere bacteria that can be cultured is oil and saline-alkalisoil(5g/kg), saline-alkali soil(10g/kg), farmland and saline-alkali soil orderly, whereaschange of the number of rhizosphere actinomycetes and fungal that can be cultured isfarmland, saline-alkali soil, oil and saline-alkali soil(5g/kg) and saline-alkali soil(10g/kg)orderly.2. BIOLOG results showed that during the whole growth period of alfalfa andtriticale, the changes of AWCD and microbial community functional diversity index(Shannon index, Shannon index, Simpson index, Mcintosh index) are farmland,saline-alkali soil, oil and saline-alkali soil(5g/kg) and saline-alkali soil(10g/kg) orderly.PCA analysis showed that the difference between farmland and oil and saline-alkali soilwas significant in all periods.3. DGGE fingerprint analysis results showed that the bands of oil conducted soilsamples were more abundant, and the diversity and uniformity of which increased.According to16S rDNA analysis, the eleven bands of alfalfa results were showed below:bands3,4,8,9,10were identified as Acidobacteria sp., bands2,7were identified asPseudomonadaceae sp., bands6,11were identified as Gamma proteobacterium sp., bands1,5were identified as Uncultured bacterium, the eight bands of triticale results wereshowed below: bands1,2,3were identified as Acidobacteria sp., band4were identifiedas Uncultured bacterium, bands5were identified as Pseudomonadaceae sp..4. After three growth stages of alfalfa and triticale, determine the oil degrade rate. The oil degrade rate of5g/kg was higher than that of10g/kg for both alfalfa and triticale,and the difference was significant (P<0.05). The oil degrade rate of alfalfa was higherthan triticale for the same conduction, and the difference was significant (P<0.05).
