| The PsaH gene was one subunit of photosystem â… , which mainly existed in green algae and higherplants and other eukaryotes, and it played an important role in Photosynthesis. The PsaH gene was clonedfrom the corn, named ZmPsaH(photosystem â… reaction center subunit H). The open reading frame was429bp, encoding143amino acid protein with a predicted molecular weight of10.2kD and a pâ… of7.30.Analysis of phylogenetic indicated that the ZmPsaH gene had high protein homology with Oryza sativa,Sorghum bicolor and Hordeum vulgare. The Real-time Quantitative PCR assay revealed that the ZmPsaHwas induced by salt and dehydration. The ZmPsaH gene was inserted into eukaryotic expression vectorpREP-5N, through transformating into Schizosaccharomyces pombe and carrying on resistance functionidentification. It simulated of salt and drought stress with NaCl and KCl, Sorbitol, respectively. Therecombinant yeast strains can present good resistance in600mM NaClã€1.4M KClã€3.0M Sorbitol culturemedium. Analysis of stress resistance suggested that the transgenic BY2cells, which havepCAMBâ… A2300-ZmPsaH Carrier, can show good resistance under salt and osmotic stress conditions.Therefore, it can be inferred that the overexpressions of the ZmPsaH gene can significantly improve stresstolerance of plant in high salinity and dehydration. |
