The research group studies on the antibacterial molecular mechanism of marigold spinosad,but we do not find relevant reports.we has obtained the resistant strain by chemical mutagenesis and screening,to lay the foundation for research marigold spinosad antibacterial mechanism in the molecular level.This test used diethyl sulfate on Fusarium spore suspension for mutagenic treatment under different times,screening out of a drug-resistant strain.Resistant strain for the test material was to extract DNA via using the method of SDS-CTAB,at the same time,we researched the mutation sites of anti-fungicide strain.At the comparison of CK,the biological characteristics of the mutant were studied.The main findings are as follows:First,we successfully obtain the anti-fungicide strain.the genetic characteristic of the resistent is stable,the growth of radial mycellium is vigorous.Second,there are several mutations in the gene of the mutant.The mutation sites of the drug-resistant strain suggest that,on the one hand,there is a mutation site in the chitinase gene fragment,and bioinformatic analysis reveals that the site encoding amino acids locates in the a-helix,at the other hand, the cellulase gene fragment,there is a mutation, it is with homology of the glycoside hydrolase family45of Gibberella zeae.Third,compared with the untreated strain,the biological characters of the mutant are mutated.The growth rate of the mutant is significantly faster,but the accumulation of the mycellium dry matter is low,the ability of asexual reproduction do not decrease,it has a good adaptability to different temperature,pH and osmotic pressure.The study has been screened of the mutant by chemical mutagenesis. And it is to lay the foundation for revealing the sterilization mechanism of marigold spinosad in the molecular level.
