Fusarium is a kind of common fungus with varieties. It’s widespreading and rich in resources. The test was separated and purified121strains of Fusarium from different hosts and different geographical locations. We used the SDS-CTAB Genome Drawing Method and Molecule Marking Technology. The inherited diversity of the fungus were analyzed after it’s amplification, the results are as below:1. the genome is more integrated.by using the SDS-CTAB Genome Drawing Method and has a higher qualityand was tested by0.8%AGE (agarose gel electrophoresis).2. There are25pairs primers from which Li and Quiros recommened and later generations complemented, in the SRAP-PCR amplification system,the renaturation variable temperature methed was used to select the12primers which have more brighter distinct stripe and polymorphism obviously..12primers were screened to amplify DNA of121Fusarium isolate.The result showed that it was amplified at a total number of260fragments using the SRAP-PCR amplification system, which were located between100-1500bp. The number of the Polymorphic loci were260andthe polymorphism rate reached100%.For each primer,the number of amplified fragment was between19-30,and each pair of primers amplified21.6fragements on average.3. The cluster analysis on the result of121strains of Fusarium’s SRAP-PCR,121strains of Fusarium can be divided into six types on the condition that the inheritance similarity factor is0.814. Six groups in the different genetic similarity coefficient can be divided into12sub-groups. reflected througt the results of fragment atlas amplification reflected that the Fusarium genetic diversity coincided with theClassical Taxonomy Morphology,which used the the SRAP-PCRA molecular technology.4. Selected the different species of Fusarium, same specie from different hosts in different location, same specie from same hosts in the different loctation, same specie from different hosts in the same loctation, and same specie from same hosts in the same loctation, and analyzed them with the genetic diversity.The results showed that different species of Fusarium have more genetic diversity than others and contain abundant hereditary variation.for the different strains of Fusarium in same species, the genetic similarity coefficient from different strains of same species is no correlation with the host and the location.5. There is a clear fingerprinting about121strains of Fusarium based on12pairs of primers with the sequence-related amplified polymorphism technology, and every isolate had a unique fingerprinting, and we can distinguish different species of Fusarium directly. The result showed that the fingerprinting of the SRAP technology is feasible to identified the different species.
