To explore the epidemiological Status of Newcastle disease virus (NDV) in chicken, five NDV strains isolated from outbreaks in chicken flocks in Shanxi Province during the years2011-2012were analyzed for the molecular epidemiology in the present studies. According to the F gene sequence of NDV strains in GenBank (FJ969394), primer was designed and synthesized. The F genes of isolated strains were amplificated with specific primers by RT-PCR, then cloned into PMD19-simple T vectors, certified by PCR and sequenced. By using DNAstar7.1software, Comparison has been made on the nucleotides of the gene F and their deduced amino acid sequence with represent NDV strains. The genotypes of the five strains have been established by Phylogenetic tree.By analyzed the sequence of the F gene, four strains belonged to genotype Ⅱ, which homology of nucleotide sequence were up to99.2%-99.4%and amino acid sequence were100%compared with LaSota strain. the cleavage site was composed of112G-R-Q-G-R-L117, which revealed that these4strains were attenuated strains. The homology of nucleotide and amino acid sequence of the other strain were95.6%and97.5%, shared with JS/5/01/Go strain which belonged to genotype VIId. the cleavage site of this isolated strain was composed of112R-R-Q-K-R-F117and have structure of K101andV121, which revealed that it was velogenic strain.
