The study on the basis of graphic groups through3options which wild germplasm sugarcane Saccharum Spontaneum L. and analysis of the tillering rate, variation of sugar content, fitting the normal distribution analysis and molecular polymorphism analysis, established the mapping population GXS85-30×GXS87-16combination for construction of the genetic linkage maps. Then choosed materials were F1true hybrid population of GXS85-30×GXS87-16, the molecule genetic linkage map of Saccharum Spontaneum L. was constructed by SSR, It will lay the good foundation looking for sugar cane some useful genetic resources. The main results were summarized as follows:1. To determine the genetic mapping was used to5th combination separate groups tillering rate variation amplitude was495.8%, variation coefficient was33.1%, the stem sugar variation amplitude was4.0%(absolute value), the variation coefficient was26.7%.2. Screened out110SSR primer pairs from414SSR primer pairs, which could show better repeatability and high polymorphism between GXS85-30and GXS87-16.3. Using that110primers analysis the polymorphism of mapping populations, in the final, there are99primers mapping populations of amplified polymorphic generated279separation markers.4. A chi-square test was performed on the279separation markers, it contain136single dose markers,32double dose markers, there are111segregation, the partial separation rate is39.78%.5. Integrating other experimental group of100single double dose segregating markers AFLP,44single double dose SCoT with this study of168SSR segregating markers. Accounting by Disto software analysis of single and double dose bands. The results show that,54SSR markers,32AFLP markers,6SCoT markers for a total of92marker loci into the map, which distributed in30linkage groups, covering the total length of map is1182.29cM, with an average distance of12.99cM between markers. |
