Northeast forest region is China’s largest natural forest, Pinus koraiensis, Phellodendron amurense and Juglans mandshurica are valuable tree species of the northeast China, as the genetic diversity of the three valuable tree species in northeast China is unclear, this situation seriously affect the protection germplasm resources and genetic improvement of the valuable timber species. This study application of SSR technology research genetic diversity of the81populations of Pinus koraiensis,30populations of Phellodendron amurense and17populations of Juglans mandshurica in northeast China. The purpose is to reveal the level of genetic diversity and provide a theoretical basis for its rational utilization and effective protection. The researchful results as follows:1. Thirteen SSR polymorphism primers of Pinus koraiensis had vivid amplification band and abundant polymorphism had been screened using magnetic beads enrichment method. Fifteen SSR polymorphism primers of Phellodendron amurense had vivid amplification band and abundant polymorphism had been screened. Twenty SSR polymorphism primers of Juglans mandshurica had vivid amplification band and abundant polymorphism had been screened.2. Using the self-developed SSR polymorphism primers to study the genetic diversity of Pinus koraiensis, Phellodendron amurense and Juglans mandshurica, test results show that Nei’s index of Pinus koraiensis is0.2622, and the Shannon’s index is0.5369. The result shows that Pinus koraiensis with high levels of genetic diversity in this region:Nei’s index of Phellodendron amurense is0.1531, and the Shannon’s index is0.4159; Nei’s index of Juglans mandshurica is0.1774, and the Shannon’s index is0.5017.3. The coefficient of gene differentiation of Pinus koraiensis is0.3785, genetic diversity between populations possessed37.85%. It is conclude that genetic diversity of Pinus koraiensis mostly exit in populations; Nm is1.24which means that the gene flow is nature. However, it is not very high. The coefficient of gene differentiation of Phellodendron amurense is0.1311, A higher level of genetic differentiation was detected among populations of Phellodendron amurense, genetic diversity between populations possessed13.11%, Nm is1.5836; The coefficient of gene differentiation of Juglans mandshurica is0.1039, genetic diversity between populations possessed10.39%.4. This study application of SSR technology research genetic diversity of the81populations of Pinus koraiensis,30populations of Phellodendron amurense and17populations of Juglans mandshurica in northeast China, the results show that the level of genetic diversity of Pinus koraiensis, Phellodendron amurense and Juglans mandshurica from zhangguangcai mountains and Lesser Khingan Mountains is higher than other regions in northeast China. The regions may be the refuge for natural Pinus koraiensis, Phellodendron amurense and Juglans mandshurica in Lesser Khingan Mountains and zhangguangcai mountains when Quaternary Ice Age come. According to the study of genetic diversity of Pinus koraiensis, Phellodendron amurense, Juglans mandshurica and based on the DIVA-GIS software,conservation measures are suggested.
