Puccinellia tenuiflora is a perennial herbaceous plant species, which belongs to the genus Puccinellia in Gramineae. It is not only a good forage grass, but also a good model to study the mechanism of salt-tolerance. Calli is a mass of undifferentiated cells mainly consisted of living parenchyma cells. Calli has the potential of dedifferentiation and rdifferentiation. Thus, it is a good material for cell biology, plant embryology, developmental and molecular biology. We analyzed the physiological and protomics changes in calli from Puccinellia tebyufkira. The main results include (1) the contents of proline and malondialdehyde were all increased in the salt stressed calli;(2) the activity of superoxide dismutase (SOD) and peroxidase (POD) were decreased under50mM NaCl. but increased under150mM NaCl. However, the glutathione reductase (GR) activity was increased with the salt concentration increased, but the catalase (CAT) activity was decreased:(3) we found77salt-responsive protein spots on2DE gel from the calli of Puccinellia tenuiflora. Among these protein spots.37spots were induced and40spots were reduced under50mM NaCl. Moreover,31spots were increased and46spots were decreased under150mM. Our results show that membrane system in calli of Puccinellia tenuiflora has been damaged under50mM and150mM NaCl for28days.The calli of Puccinellia tenuiflora regulated the osmotic balance by accumulation of proline. In addition, the calli triggered antioxidant defense system in order to scavenge ROS. The salt-responsive proteins implied that the calli of Puccinellia tenuiflora may enhance salt tolerant ability by regulating numerous metabolism pathways.
