The utilization of heterosis and the research of the mechanism of male sterility in wheat has been the research hot spot in genetic breeding for many years. Wheat male sterile can be divided in genetic male sterility and. Cytoplasmic male sterility (CMS) is caused by the extranuclear genome (mitochondria or chloroplast) and shows maternal inheritance, and physiological male sterility is induced by chemical hybridization agent. The research about wheat male sterility mechanism has quite a few reports, but so far the complexity of the sterility genes has not obtained substantial progress.The experiment use the technology of paraffin wax slice to observed the development of anther wall and pollen, which materials are normal fertile Xinong1376and cytoplasmic male sterility wheat Xinong1376. Compare the two materials to get the cytological differences and their respective characteristics. Through TUNEL detection, we can observe the tapetum PCD process and its relationship with the pollen abortion. In addition, we use agarose gel electrophoresis to detect the DNA ladder differences between the materials. We can confirm the abnormal phenomenon of the male sterility on the cytology and molecular biology level, and lay the foundation for to reveal the physiological mechanism of wheat male sterility.The important results obtained are as follows:1) In the early mononuclear, the results is no differences between two kinds of materials, including aceto-carmine staining, TUNEL fluorescent staining and DNA ladder detection of sterile and fertile wheat anthers.2) In the late mononuclear, sterile and fertile wheat anthers began to appear differences. In the sterile wheat anthers, the microspore nuclear of aceto-carmine staining appear indistinct, the fragment of nuclear DNA by agarose gel electrophoresis and TUNEL-positive signal in tapetum are detected. But tapetum degradation is finish and the fertile wheat microspore is normal development.3) In the tninuclear stage and bninuclear stage, the microspore of sterile wheat anthers detect more fragment of nuclear DNA than the late mononuclear and nuclear of aceto-carmine staining appear indistinct, besides TUNEL-positive signal in microspore nuclear are detected. But DNA fragment and TUNEL-positive signal are not detected in fertile wheat anthers.4) The time of tapetum degradation is delayed in the sterile wheat anthers form the detected of the TUNEL fluorescent staining, in addtion TUNEL-positive signal is also can be seen in the microspore of sterile wheat anthers, The results show a close relation to the pollen abortion.
