| DHAV-1(Duck hepatitis A virus type1) causes a highly contagious disease in ducklings, characterized primarily by liver necrosis, hemorrhages, and high mortality. The disease can cause mortality up to95%in young ducklings during the first week of life and economic importance to all duck raising farms because of the high mortality. At present, the research of duck hepatitis is mainly focus on sequeneing the genome of DHAV-1and diagnosis and controling of DHAV-1and so on, less on pathogenic mechanism of virus, the virus pathogenic mechanisms are still not very clear. In this study, on based of the establishment of viral infection model, we investigated differentially expressed genes in the infection process, screening the viral infection associated gene of duck by RNA-Seq technology. This paper studies the results are as follows:4-day-old were expermentally infected with DHAV-1from intramascular infection, the pathological model of duck virus hepatitis has copied successsfully, by histopathology and viral PCR detection method. The liver and spleen of the ducklings were collected, and extracted tissue RNA by RNA-Seq. Analysised and processed the data, the results were that liver and spleen of the infected ducklings compared with those of the control group, a total of7599genes were differentially in the liver and spleen,3521genes upregulated and4078genes downregulated. The expression of the genes in both liver and spleen of the infected ducklings and those of the control group were categorized by hierarchical clustering with average linkage measurement. The most up-regulated gene clusters came together, the down-regulated genes distribution is relatively scattered. The application of fluorescent quantitative PCR method and semi-quantitative PCR method validation analysised the differential genes of RNA-Seq, the results showed that the genes m-RNA expression levels changed differentially expressed in multiples and expression profiling sequencing results, which showed that the sequencing data was the true and reliable.The conclusion were that DHAV-1may through partial metabolism gene down-relegulated, suppressing host cell metabolism, causing many metabolism intermediary product stored, after ducklings infected DHAV-1.Ducklings may through partial immunity gene up-relegulated, suppre-ssing virus’s duplication in cell, which lay the foundation for further studied the pathogenesis mechanism of DHAV-1. |
