To explore the early action mechanism of Znonotus obliquus polysaccharides in the treatment of mice infection acutely with Toxoplasma gondii and AP-1signaling pathway, after buliding the model of mice infected acutely with toxoplasma gondii, we divided it into negative group, tested group and treatment group randomly. Using ELISA to detect the serum levels of TNF-β、L-6、IL-1β and IL-12, RT-PCR assay to detect the expression of mRNA of c-Jun and IL-1β and IL-12in the liver tissue of spleen and to detect the protein expression of nuclear transcription factor c-Jun by western blot.The results of ELISA showed that the expression of IL-12, IL-6, IL-1β and TNF-α in both the RH strain infected group and PLK strain infected group were increasing significantly, and reached the peak at34h,48h,24h,24h,respectively, then showed downward trend. The expression of IL-12, IL-6, IL-1β and TNF-α in the RH strain infected group were higher than the PLK strain infected group at12h to60h. The expression of the two tested group during12h to60h were higher than the negative group, but lower than the model group. The differeirces between the RH strain infected group and the tested group were more significant than the differences between PLK strain infected group and the tested group (P<0.01). Results showed that the expression of IL-12, IL-6, IL-1β and TNF-α in the RH strain infected group were higher than the PLK strain infected group, and Znonotus obliquus polysaccharides could inhibit the expression of IL-12, IL-6, IL-1β and TNF-α in the infection of RH strain infected group and PLK strain infected group. RT-PCR results showed that infected with the RH strain and the PLK strain, the IL-1β and IL-12of spleen and c-Jun of liver tissue in the negative group, the model group and the experimental group were amplified single electrophoretic bands in736bp,978bp and360bp, respectively. The electrophoretic band of the model group was the brightest, followed by the experimental group and the negative group. Gray-scale analysis results showed that, there was significant difference in the infected group with the expression of IL-1β and IL-12in spleen and c-Jun in liver tissue compared with the model group (P<0.01).The tested group and model group were significant difference from the negative group (P<0.01). The expression of IL-12in the PLK strain infected group was no difference compared with the RH’s (P>0.05), while the expression of IL-1β andc-Jun in the RH strain infected group was significantly increased compared with the PLK’s (0.01<P<0.05).Results showed that mice both infected with RH strain and the PLK strain, the IL-1β and IL-12of spleen and c-Jun of liver tissue expressed significantly and the expression of IL-1β and c-Jun in RH strain infected group was higher than PLK’s. Znonotus obliquus polysaccharides inhibited transition releases of IL-1β and IL-12of spleen and c-Jun of liver tissue in both the RH strain infected group and PLK strain infected group. SDS results showed that c-Jun(p39) in liver tissue of the two infected groups, the negative group and the model group were consistent with the target protein bands at39KD. The model group was the brightest, followed by the experimental group and the negative group. Western blot results showed that compared with the PLK strain, the protein of c-Jun in RH strain was much wider and the color was much deeper. Gray-scale analysis showed that the difference between the RH strain infceted group and the PLK suain infected group was significant (P<0.01). Results showed that Znonotus obliquus polysaccharides could inhibit the transition releases of c-Jun protein in both the RH strain infected group and the PLK’s.In conclusion, Znonotus obliquus polysaccharides inhibited transition releases of IL-1β, IL-6, IL-12and TNF-αin sera of mice infected acutely with Toxoplasma gondii. It suggested that by down-regulating the mRNA of IL-1β,IL-12and c-Jun transition expression.
