Gentiana rigescens Franch. ex Hemsl. belongs to Gentiana herbs of Gentianaceae sp., reckoned among genunie medicinal materials from Yunnan province. Modern pharmacology research has shown the content of gentiamarin (C16H20O9) which is the active component of Gentian directly affects the quality of Gentian. Gentiana rigescens is one of the main wild medicine materials of the extraction of gentiamarin. Gentiana rigescens principally reproduces by seeds.In this study, Gentiana rigescens seeds are dried in the shade and stored under natural indoor conditions. Ninety days later, the seed treatment, which consist of seed priming and rehydration-dehydration, is applied to Gentiana rigescens seeds by diffferent inorganic salt solution and macromolecular organic solution. This study aims to discuss the effect of different priming and dehydration treatment on seed germination in Gentiana rigescens and to screen out optimum priming solution combined actual production circumstance by means of germination test, physiology and biochemistry test, counting and analyzing germination characteristic index(germination energy, germination percentage, germination index and vigor index) and physiological characteristics index(activity of peroxidase and content of soluble sugar, free proline and malondialdehyde) after the treatment.The main results are as follows:1. Gentiana rigescens seeds dried in the shade and stored under natural indoor conditions for ninety days, each of germination characteristic index has been at a lower level. The germinating capacity of these seeds is almost lost. Therefore, it is unsuitable to sow.2. Hydropriming-dehydration, to a certian extent, lends itself to the restoration of seed vitality but not helps much.3. Each of germination characteristic index has reached the significance level(P>0.05) after1.0%NaCl,1.0%KN03,4.0%KH2PO4,1.0%Zn(NO3)2,10%PEG-6000priming and dehydration treatment on seed germination in Gentiana rigescens. The germination effect of1.0%KNO3priming and dehydration treatment is the best among all priming solutions and the germination percentage of Gentiana rigescens reaches81.67%. Compared to CK1, the germination index and vigor index of Gentiana rigescens respectively increase to34times and248times.4. Compared to CK1, CK2and other treatments, the sprouting time of Gentiana rigescens after priming solutions(containing chloride ions) priming and dehydration treatment is advanced.5. Compared to CK1, CK2and other treatments, the radicle length and seed vitality of Gentiana rigescens after priming solutions(containing nitrate ions) priming and dehydration treatment are advanced. The germination percentage of Gentiana rigescens reaches75.00%after Zn(NO3)2priming and dehydration treatment. Compared to CK1, the germination index and vigor index of Gentiana rigescens respectively increase to31times and244times.6. Compared to CK1, CK2and other treatments, the sprouting phase of Gentiana rigescens after priming solutions(containing nitrate ions and PEG-6000) priming and dehydration treatment is advanced.7. It is propitious to accumulate soluble sugar and free proline, remove malondialdehyde and increase the activity of peroxidase in seed cell after priming and dehydration treatment in Gentiana rigescens.8. The effect of the accumulation of soluble sugar and free proline after10%PEG-6000priming and dehydration treatment in Gentiana rigescens is the best.9. The effect of the removal of malondialdehyde and the increment of the activity of peroxidase after1.0%KNO3priming and dehydration treatment in Gentiana rigescens is the best.10. The major factor totally promoting germination percentage is the removal of malondialdehyde and the high activity of antioxidant enzyme in seed cell after priming and dehydration treatment in Gentiana rigescens, with the accumulation of soluble sugar and free proline playing a less role in totally promoting germination capacity.It is concluded that1.0%KNO3priming and dehydration treatment in Gentiana rigescens can gain much higher productive result in actual production before sowing.
