In order to find out the types, community structure and ecological distribution of Pu lsatillae Radix (which belongs to medicinal plant), and further to screen the endophytic f ungi which can be used in pollution-free plant disease prevention and control of resources, five Chinese bulbul sample area were chosen in Xinyang and Zhumadian cit y of Henan provinence acording the different ecological environments. In this study, the distribution and species diversity of Pulsatillae Radix’s endophytic, and the interaction of asntagonistic fungi and pathogenic fungi were researched by picking, separating, identify ing, antagonistic experiments and biochemical analysis. The results indicated that Pulsatil lae Radix’s endophytic have more species-rich, widely distribution, and biodiversity. The re were totally25species of endophytic fungi in14genera belonging to8family, and fiv e kinds of Endophytic fungi (trichoderma, penicillium, quasi stem point mould genera, bi g stem point mould genera and alternaria bacteria genera) were the dominant species, whi ch have a wide distribution in different seasons and higher separation rates. Furthermore, the quantity of endophytic fungi are different in different tissues and organs, and compari ng the spring, the numbers and kinds of endophyti-c fungi were more in autumn.The different pathogenic fungi were used to study the antimicrobial resistance of paulsrtilla chinensis. Three strong antagonistic strains were identified after two standoff screening. The result showed that the XY25strain has broad-spectrum resistance and strong inhibitory effect on the fruit rot fungus. The XY25strain belongs to the genus Penicillium fungi after morphological and molecular identification. The optimum growth temperature of XY25strain was30℃to35℃, and the optimum growth pH was7.0to7.5. The study on the XY25reproduction rate with different carbon source and nitrogen source illustrated that the optimal carbon source and nitrogen source were glucose and peptone, respectively.Hyphostroma and liquid fermentation isolated when fermenting in liquid culture medium can inhibit the growth of disease spot and reduce the rate of fruit rot. The result indicated that the mycelium extract contains antibacterial ingredient. Thin-layer chromatography was used to analysis the mycelium extract, the result showed that mycelium extract contains five component, especially component I had an inhibitory effect on pathogenic bacteria growth. Therefore, the XY25strain of endophytic fungi of paulsrtilla chinensis has potential application value. It has a strong broad-spectrum antibacterial property on plant pathogens, because the mycelium cells can produce antimicrobial substances during growth and development process.
