| The genetic structure of3strains of Mongolia Dogs was investigated by microsatellite analysis. A molecular phylogeny study was conducted to reveal the origination of Mongolia Dogs by using mitochondrial D-loop analysis.Experiment1. Microsateilite Analysis of Three Strains of Mongolia DogsPolymorphisms of eight microsateilites were analyzed in three strains Mongolia dogs with different feather color. The results showed that8microsateilite loci were high polymorphic in three strains of mongolia dogs. Polymorphism information content (PIC) and heterozygosity(H) were0.5030-0.6171,0.5614-0.6770, respectively. The coefficient of the three strains was0.2183, which indicated there were major medium differentiations among these three strains. The migration rate ranged from0.4532to1.4178, with the average of1.0234. Our result suggested that there was genetic correlation among the three strains, which might reflect the history of the breed origination.Experiment2. Phylogenic analysis of Mongolia Dogs by comparing D-loop of mitochondrial DNA.In this study, primers were designed according to the mitochondrial genome sequence of domestic dog to get the full D-loop sequence of Mongolia Dogs. The phylogenetic trees of Mongolia Dog and16representative breeds were constructed using grey wolf and coyote as outgroup. The results showed that, the D-loop region of Mongolia Dog was1278bp, in which there existed10bp-repeat units with the core sequence of TACACGT(A/G) CG. There were31repeat units in Mongolia Dogs, which was one more repeat unit than in Tibetan mastiff. The content of A+T was more than that of G+C in D-Loop sequence of r Mongolia Dogs. Phylogenetic analysis revealed that, Mongolia Dog, grey wolf, Flat Coated Retriever, Poodle, Shetland Sheepdog and Saint Bernard were clustered together. This confirmed the viewpoint that Mongolia Dogs originated from grey wolf, and the fore-mentioned breeds might have similar genetic origination with that of Mongolia Dogs. |
