| Bovine mastitis is one of the most common diseases of dairy cow breeding, cause huge economic losses, which is influenced by many factors, of which the pathogenic microorganisms infection is one of the main.Experimental objective:This experiment from actually, combined with clinical practice, collecting Milk sample from a dairy farm.Counting on milk somatic cell count and differential counting, The separation of bacterial culture, identification and drug susceptibility test.Experimental results of the data then fed back to cow, guide the clinical medication and breeding management.Experimental methods:Using somatic cell count meter somatic cell count on milk samples with Newman’s dyeing method, somatic cell count under the microscope.Willing milk samples were inoculated on blood AGAR medium, QueFuMan and MaiKangKai culture medium for separation of the bacteria culture.Cultivating the typical biochemical identification of bacteria, staphylococcus aureus five biochemical identification of genus,14biochemical identification of E. Coli. Identificated the bacteria respectively made21kinds of drug susceptibility test.The experimental results:Collecting the80cases of milk samples, somatic cell count in under2x105are34cases, between the2x105-5x105are9cases and more than5x105are37cases. The incidence rate of mastitis is57.5%, the Subclinic mastitis incidence rate is11.25%, The Subclinic mastitis take mastitis incidence rate is about19.57%Somatic cell mainly neutrophils, macrophages and lymphocytes, and the three cell accounts for about98%of the total number of somatic cells. The macrophages are the mainly cells in normal milk and the neutrophils are the mainly cells in mastitis milk.Cultivate the bacteria are mainly composed of staphylococcus aureus and E.coli, not cultivate streptococcus. Culture bacteria in43cases of milk samples, Staphylococcus accounted for69.77%, Enterobacter accounted for30.23%. Staphylococcus aureus accounted for44.19%, epidermis Staphylococcus accounted for18.60%, Staphylococcus saprophytic accounted for6.98%, Escherichia coli accounted for13.95%, Proteus sp accounted for9.30%, Shigella Chalmer accounted for4.65%, Klebsiella sp accounted for2.33%. To identify the bacteria made21kinds of drug sensitivity test, to ciprofloxacin, levofloxacin, streptomycin, amikacin, kanamycin and has a high sensitivity of gentamicin.The drug resistance of gram-negative bacteria than gram-positive bacteria, all strains resistant to penicillin drugs.Experimental results:The main somatic cells in milk are neutrophils〠macrophages and lymphocytes.
