In the present study, we detected the genetics diversity of HSP70and HSP90genes among Fast Partridge, D-Dwarf chickens, Guangxi local breed2chickens, Silkies and lingshan chicken, compared the genotype distribution and analyzed the association between the polymorphisms and carcass traits in different breeds. At the same time, we analyzed HSP70and HSP90gene’s expression in differsnt tissues of chickens by real-time fluorescence quantitative PCR. The results are as follows:1. The relationship between polymorphisms of HSP70gene and slaughter traits of chickensThree genotypes(AA/GG/AG) of HSP70gene were detected using SB-ASA in chickens. Association analysis revealed that the heterozygous(AG) birds had significant higher BW, BP than homozygous(GG) individuals in male Partridges. The OW, CW, HW, EW, CP, BW, BP had significant difference in D-Dwarf male chickens. The homozygous(GG) birds had significant higher HW, LP than heterozygous(AG) and the homozygous(AA) birds had significant higher AW than the homozygous(GG) ones in D-Dwarf female chickens. It had significant difference between AA, GG and AG in CP of Guangxi local breed2female chickens, BP of male Silkies, CP of female Silkies. The heterozygous(AG) birds had significant higher CW, HP, EP, EW, LW than homozygous(GG) in lingshan male chickens. The CP had significant difference in lingshan female chickens.2. The correlationship between genotypes of HSP90gene and slaughter traits of chickensThe mutation of T→C happened on exon2of the HSP90gene lead to a substitution of the amino acid Phe for Ser. was detected using PCR-RFLP, SB-ASA and sequencing technologies. There were two genotypes:TT and TC. By the least square means(LSM), the homozygous(TT) birds had significant higher BW than heterozygote(TC) in D-Dwarf male chickens and the heterozygous(TC) birds had significant higher BP than homozygous(TT) in lingshan male chickens. In the other breeds, it had no significant difference in the carcass traits. 3. Expression profile of HSP70and HSP90genes in chickenExpression patterns of HSP70and HSP90genes in the thirteen tissues were finished by real-time fluorescent quantitative PCR in chickens. The results showed that HSP70and HSP90genes expressed in all the tissues detected, but the expression levels were quite different. HSP70gene’s expression in Pancreas, leg muscle and abdominal fat was the most, followed by kidney, skeletal muscle, heart, liver, spleen, cerebellum, hypothalamus and pituitary, and lowest in lung and brain. HSP90gene’s expression in heart, cerebellum, pituitary, leg muscle, abdominal fat and skeletal muscle is the most, followed by liver, pancreas, brain, hypothalamus and abdominal fat, lowest in spleen, lung and kidney. |
