Lentiviral Vector Mediated IGF2B Gene Over Expression In Dorsol Muscle Of Common Carp And The Effect On Growth Of Common Carp(Cyprimus Carpio L.)

Insulin-like growth factor2(IGF2) is a growth factor that could promote cell division, there were two kinds of IGF2gene in common carp, insulin-like growth factor2a (IGF2a) gene and insulin-like growth factor2b (IGF2b) gene. In order to know more about the effect of insulin-like growth factor2b(IGF2b) on the growth of common carp, dorsol muscle of common carp were mediated by Lentiviral Vector containing enhance green fluorescent protein (EGFP) gene and IGF2b gene by the dose of10μl、20μl and40μl to observe the growth situation of experiment groups mediated by Lentiviral Vector.250Jian carps that were nearly same were chosed for experiment. The fish wrere randomly divided into five groups, and the first group was used as control group, the second group was injected with20μl water, the third group、forth group and the fifth groups were respectively injected with10ll、20μl、40μl Lentiviral Vector that include IGF2b gene and EGFP gene (the third group、forth group and the fifth group were all called experimrnt groups). The fish would been cultured in aquarium for48hours for observation, then were respectively shifted to five cages of a same pond, and were regularly fed with same amount of same feed every day.48hours later, five fishes of experiment groups were randomly chosed to examine the expression of IGF2b gene by observing if there was green fluorescent and the range of them. Four months later, body weight、body length body height and body thickness of common carp were examined, at same time, concentration of serum IGF2、IGF2R、IGF1R and serum alanine aminotransferase(ALT)、 alkaline phosphatase(ALP)、blood urea nitrogen(BUN)、creatinine(CREA)、total protein(TP、total cholesterol(TC) were examined to observe the effect of overexpression of IGF2b gene on growth and serum parameter of common carp. At last, real time PCR was used to observe the express situation of IGF2b gene in dorsol muscle of common carp.The result showed that green fluorescent were observed in dorsol muscle of common carp belonging to experiment groups forty-eight hours later, so IGF2b gene have integrated into the genome of the common carp dorsol muscle. And body weight、body length、body height and body thickness of experiment groups were significantly higher than control group (p<0.05) four months later, but water injected group was significantly lower than control group (p<0.05). Serum parameters of common carp were examined, the result showed that there were no significant difference between groups about the concentration of serum IGF2、IGF2R、IGF1R. ALT of water injected group was significantly higher than control group, and ALT、ALP、TP and BUN of10μl injected group were significantly higher than control group (p<0.05), and serum parameters of20μl injected group and40μl injected group both had no significant difference with control group(p>0.05). At same time, the result that IGF2b gene were successfully overexpressed in dorsol muscle of experiment groups was examined by real time PCR, but the IGF2b gene expression of water injected group was significant lower than control group (p<0.05). So, the overexpression of IGF2b gene in dorsol muscle of common carp improved its growth, and a certain dose of Lentiviral Vector containing IGF2b gene had no significant influence on serum of common carp, but the method of injection that was used to shift Lentiviral Vector into the dorsol muscle of common carp had a bad effect on the growth of common carp and their health.
