| Porcine Circovirus serotype2(PCV2) is the pathogen of postweaning multisystemic wasting syndrome (PMWS) and, found then, that of Porcine dermatitis and nephropathy syndrome (PDNS), Porcine respiratory disease complex (PRDC), Congenital tremor (CT), etc. it has brought much economic losses to the swine industry. The genome of PCV2contains two major open reading frames, ORF1and ORF2, which enchode Rep protein related with viral replication and main structure Cap protein, respectively. Cap protein can induce protective immnunity against PCV2infection in swine for it contains several immunodominant epitopes. In this study, recombinant Cap protein was expressed in BL21to develop the indirect ELISA kit for detecting antibody for PCV2. Rabbit and mouse polyclonal antibodies for recombinant Cap protein were also produced to develop Colloidal Gold Immuno-Chromatographyic Strips for PCV2. This study mainly contains the following two parts:1Cap gene of PCV2was expressed in BL21and the recombinant protein can be used to establish the indirect kit as the antigen.2After conditions improved, indirect ELISA test method was established with the purified protein as antigen. This method has no crossing with CSF, PRRSV, PPV, etc.3Indirect ELISA kit for PCV2was developed based on the established ELISA method and this kit can used in epidemic investigation and clinical diagnosis of PCV2. The test results of serum samples from Anhui and Jiangsu should attract our attention on PCV2in these regions.4Rabbit and ICR mouse were immunised with purified recombinant protein to produce polyclonal antibodies. These two purified polyclonal Abs were tested to be much immunological and could be used to develop Colloidal Gold Immuno-Chromatographyic Strips for PCV2.5Colloidal Gold Immuno-Chromatographyic Strips for PCV2was developed with these purified two polyclonal antibodies and was tested to be qualified. This method is convenient, simple and of low coss and could be promoted clinically. |
