Porcine reproductive and respiratory syndrome(PRRS) is caused by porcine reproductive and respiratory syndrome virus(PRRSV), which is a viral infectious disease.The interaction between PRRSV and host immune responses in alveolar macrophages is the cause of the outbreak of PRRS. In order to understand the effects of PRRSV on piglet immune system, the dynamic changes of immune related genes in piglets’ porcine alveolus macrophage (PAMs) were dectected in this study.FiveSix thirty-five-day conventional post-weaning piglets were chosen as the experiment animals, which were free of PCV2and PRRSV antibody and antigen detected by ELIS A and RT-PCR. PAMs obtained from piglets were randomly divided into sixfour groups (0h attack,0h control,12h attack,12h control,24h attack,24h control). To analyse the effects of PRRSV on piglet PAMs’ innate immune, the dynamics transcription changes of immune correlation factorscytokines, include (MHC-1, IL-2, IL-6, IL-8, IL-10, SLA-1and TNF-α) were detected by Real time-PCR after extracted PAMs’ total RNA.of PAMs. The results showed that:the expressions of IL-2and SLA mRNA were significantly down-regulated at the point of24h after inoculated by PRRSV compared with control group; the expressions of IL-6, MHC-1and IL-10mRNA was markedly up-regulated at the point of24h after inoculated by PRRSV compared with control group; the expressions of IL-8mRNA were significantly down-regulated at12h and24hthe two time points after inoculated by PRRSV compared with control group; and the expressions of TNF-α mRNA were visibly up-regulated at12h and24h after inoculated by PRRSVthe two time points. The results demonstrated that after PRRSV infection, the PAMsbody appeared to severely immune suppression after a transient immune enhancement after PRRSV infection,.Five thirty-day conventional post-weaning piglets were chosen as the experiment animals, which were free of PCV2and PRRSV antibody and antigen detected by ELISA and RT-PCR. CellThe supernatants were harvested after treated with or without PRRSV for0,12, and24h, respectively. To analyse the effects of PRRSV on immune system, the expression changes of IL-6, IL-10and TNF-α were detected by ELISA. The results showed that compared with the control group, the expressions of IL-6and TNF-a were significantly increased (p<0.05) at two time points (12h and24h) in PRRSV groups compared with the control group. The expression of IL-10in PRRSV group was isignificantly increased significantly (p<0.05) at12h compared with control group;, however there was no significantly changes at24h(p>0.05). The results demonstrated that PRRSV could activate the immune response, but caused significant immune disorders at the same time.
