Infectious bovine rhinotracheitis (IBR) can cause multiple system acute contagious disease with character of latent and persistent infections. This study separated and cultured adherent cells from bovine peripheral blood and then identified them by morphological observation and flow cytometry. The proliferation mechanism of IBRV in mononuclear macrophage was detected by absolute fluorescence quantitative polymerase chain reaction (PCR); The transcriptional level of IL4, IL-6, IL-8, IL-10and TNF-α, were analyzed by relative fluorescence quantitative polymerase chain reaction (PCR) after bovine mononuclear macrophage being infected with IBRV0,6,12,24and48h in vitro. The results are as follows:(1) The cultured adhere cells separated by density gradient separation method were identified as mononuclear macrophages by morphological observation and flow cytometry.(2) After6h which IBRV infected mononuclear macrophages, IBRV grew fastest and then the grewing speed was increment is relatively slow and at96h it reached a peak value, with the time going by,the concent of IBRV began to decrease in macrophages.(3) The infection of IBRV with mononuclear macrophages can significantly decrease mRNA expression level of IL-4、IL-6and IL-8.(p<0.05)(4) The infection of IBRV with mononuclear macrophages can significantly increase mRNA expression level of IL-10and TNF-a.(p<0.05)This study analyzed the effects of IBRV on mononuclear macrophage immunological function by detecting dynamic changes of cytokines expression level, such as IL4, IL-6, IL-8, IL-10and TNF-α, so as to provide basis for further study of pathogenesis of IBR.
