SHENZHU SAN comes from classics prescription. It was used to cure pathogenicwind-cold, headache, febricity, hating cold, body ache. Its chemical compositions containedessential oil, coumarins, amino acids, terpene, alkaloid, saponin, and flavanoid. Essentialoil was its major active component. Atractylodes chinensis (DC.) Koidz. was its principaldrug. Ligusticum sinense Oliv. Ligusticum chuanxiong Hort. and Angelica dahurica. (Fisch.ex Hoffm.) Benth. et Hook. f were ministerial drugs. Notopterygium incisum Ting ex H. T.Chang and Asarum heterotropoides Fr. var. mandshuricum (Maxim.) Kitag were adjunctivedrugs. Glycyrrhiza uralensis Fisch. was messenger drug.The subject was the subject of a technology project—Innovation of naturally occurringdrugs. According to "the method of the new drugs approval" and the standard of the sixthtype of Chinese new drugs, a systematically research on found of active compound,extraction technology and quality standard. The main contents and results included:1.Every Chinese medicine was quantified, which was up to the standard of 2005 edition’sChinese pharmacopoeiaⅠ.2.Filtrating availability part by pharmacological research: Two experiment ofantagonizing influenza A virus tests and acetic acid-induced abdominal capillarypermeability test are used to definitude the availability part much more, and determineextraction method.3.Studies on Refining Process:(1) Study on rule extraction technology of SHENZHU SAN:①Researching on extractingessential oil form Atractylodes chinensis (DC.) Koidz., Ligusticum sinense Oliv. andNotopterygium incisum Ting ex H. T. Chang: With the studies of the processing history andcontemporary documents, indicated by extraction rate of essential oil, physico-character andarea of main peaks, the optimal processing technology was identified with L9 (3)4orthogonal design. The technology parameters were described as follows: The crude drugswere soaked 12 hours with twelve times water, and heated for five hours.②Researching onextracting compositions solved by alcohol from Ligusticum chuanxiong Hort., Angelicadahurica. (Fisch. ex Hoffm.) Benth. et Hook. f and Glycyrrhiza uralensis Fisch.: The optimized extracting technology is heating the crude drugs two times for one and half hourswith eight times 65% ethanol.(2) Study on Supercritical Fluid Extraction of SHENZHU SAN: The optimal processingtechnology was identified with L18 (2×37) orthogonal design. The technology parameterswere described as follows: 10 mesh’s crud drugs were extracted four hours without entrainer.The temperature of Extractive was set to 45℃. The pressure of Extractive was set to 30Mpa.(3) Definiting the extracting technology: Indicated by extraction rate of essential oil,extract yield and diversion rate of marmelosin, supercritical carbon dioxide fluid extractionwas better than rule extraction technology.(4) Resarching on Process of clathration: To increase the stability of SFE-extractions, theywere enveloped. Indicated by rate of reclaimation and envelopement, the optimal processingtechnology was identified with L9 (3)4 orthogonal design, the factors of which wereenveloping time, ratio betweenβ-CD and SFE-extraction,enveloping temperature usedseparately. The technology parameters were described as follows:wraooing up itSFEextraction:β-CD (1:6), and stirring for three hours under 40℃.(5) Resarching on driving out garbage: The optimized technique is concentratingextraction as 1.0g/ml,keeping 18 hours with making it in 60% ethanol.4. Establishing the quality standards: Atractylodes chinensis (DC.) Koidz., marmelosin,isoimperatorin and ferulaic acid were indentified by TLC. TOX ingredients of Asarumheterotropoides Fr, var. mandshuricum (Maxim.) Kitag were checked out by GC-MS. Thecontents of marmelosin was measured by HPLC.The quality standards of the crude drugsand finished products were established, the protocol was referred.
