A recently isolated microorganism, Acetobacter suboxydans 605, wasused for transformation of glycerol to dihydroxyacetone. Thetransformation performance with resting and immobilized ceils wereoptimated to improve dehydrogenase activity for industrial application.The culture and glycerol dehydrogenase production conditions ofAcetobacter suboxydans 605 were studied in detail employing glycerol assubstrate. The better results of dihydroxyacetone production were obtainedby Acetobacter suboxydans 605 resting ceils under the optimalbiotransformation conditions of temperature 30℃and pH Value 6.0 inphosphate buffer solution. Cell enzyme activity of transfromation fromglycerol to dihydroxyacetone could be significantly inhibited by NH4+ andFe3+, whereas it can be improved by Ca2+、Mg2+、Co2+. Substrateinhibition could occur when glycerol concentration reach 0.9%or above.After 5 batches, 78%enzyme activity of the resting cells was remained.The optimal biotransformation conditions by cells immobilized with sodium alginate in saline (pH 7.0) were as follows: pH 5.5, temperature35℃. Compared with free cells, the enzyme activity of immobilized cellswas more stable.
