The Coenzyme Ql0is a prenylated benzoquinone lipid, and it is also named ubiquinone.The chemical name is2,3-dimethoxy-5-methyl-6-polyprenyl-1,4-benzoquinone. Members ofthe Coenzyme Ql0are found in microorganisms, plants, and the heart, kidney, liver of animal,where they function are redox carriers in mitochondria or in association with respiratoryprocesses of electron transport chains. It is an antioxidant produced naturally in the cells.TheCoenzyme Q10exhibited excellent medical and physiological activities against variousdiseases such as heart diseases, cancer, hepatitis and so on.Mutagenesis and screening of mutant strains with high yield of Coenzyme Ql0,optimization of fermentation conditions,extraction and determination of Coenzyme Ql0werestudied in this text. Saccharomyces cerevisiae FCC2146were used in the fermentation of coenzyme Q10production at the first time.Experiment to determine the acid-acetone as a separation and extraction method ofcoenzyme Q10, it is simple equipment, energy consumption and low cost, and also lessdestructive to the coenzyme Q10.At the same time, the production of coenzyme Q10wasidentificated and determinated by combining of Thin-layer Chromatography (TLC) and HighPerformance Liquid Chromatography (HPLC).Saccharomyces cerevisiae FCC2146were investigated from five different speciesidentified in the Blastocystis,and its dry weight of thalli was4.67g/L、the yield of intrallularCoQl0was1.239mg/g and coenzyme Q10production was5.785mg/L.Optimize fermentation conditions and culture media for CoQl0production by ResponseSurface Methodology,the results showed that Glucose concentration was1.95%,yeast powder+soy peptone(1:5) concentration was0.483%,2.55%carrot juice was added as growthfactor,initial pH was6.0,the medium volume was50mL in250mL Erlenmeyer flask, inocul-um concentration was7%, the fermentation temperature was30℃,the fermentation time was48h,the yield of CoQl0reached16.4mg/L,which was183%higher than that beforeoptimization....
