Opioid peptides, a kind of anti-fatigue peptides, have a deeply effect on improvedhuman body function, relieve fatigue, ameliorate sleep situation and so on. This topic ismainly about the powder milk rich in opioid peptides. The preparations of opioid peptidesare produced by hydrolyzed casein using pepsin. Then put the debitterized hydrolysatesinto powder milk, formulated the powder milk with the effect of anti-fatigue. This researchis based on the previous study, all the former study have been determined the powder milkwith opioid peptides has the function of anti-fatigue by animal experiments. This study ismainly about: pilot test of powder milk which is rich in opioid peptides; edible test byrecruiting volunteers in order to determine the human cytokines and the questionnaire asaddition. All by above, evaluated the anti-fatigue effect in powder milk more accurate,comprehensive, and humanity.The pilot test of powder milk with opioid peptides produced about2kg enzymatichydrolysates from casein, and25L enzymolysis liquid each batch;a total of10batches.According to the optimized conditions of enzymatic hydrolysates from casein tohydrolyzed protein, conditions as following: the substrate concentration5%, enzyme isadded as25,000U/3g substrate, pH=2.5,43℃as the enzymolysis temperature. After thehydrolysis, using β-cyclodextrin (β-CD) debitterized, the condition is: the concentration ofenzymatic hydrolysates from casein is0.6%, concentration of β-CD is2.5%,40℃as thetemperature of debitterizing,30min stirring reaction. Then desalination and concentrationproduced the powder of enzymatic hydrolysates from casein by freeze drying. According tothe formulate: each100g powder of milk add4.2g enzymatic hydrolysates from casein,17gβ-CD, and mass fraction of1%sucrose to premixed opioid peptide powder milk.Research the effects to human body cellular network communication by full of opioidpeptides. Recruiting young healthy volunteers as the experiment object divided thevolunteers into3groups with: add enzymatic hydrolysates from casein group(Casomorphin); no add whole milk powder; and add opioid peptides as formula milk, eachgroup has10volunteers. Experimental need2days as total, the first day as a control case,all the volunteers take the same food, work and rest regulated, any outside food is forbidden,do not take any major physical sports.3hours after the lunch take4mL blood samples ascontrast ones. All the contrast samples should be let stand4hours in4℃, and under thesame tempters situation to centrifugal separation and repackaging, keep in-70℃for thefollowing needs. The following day, all the volunteer take the same regular of the first dayin working and rest, after the lunch, as the dividing condition instruction, take each kinds ofpowder milk separately: enzymatic hydrolysates from casein (Casomorphin)4.5g/person; whole milk powder25g/person; and powder milk rich in opioid peptides25g/person. After3hours take4mL blood samples, with the first day processing preserved under test. UsingMillipore Kit and liquid chip scanner to detect the before and after group’s concentration of38kinds of cytokines in serum, using paired sample T test method to deal with testing data.According to the result,7kinds of cytokines in cytokines have significant changes,respectively, IP-10, VEGF、IL-17A、IFN-γ、G-CSF、IL-1ra and TNFα. Results showed that: the powder milk rich in opioid peptides has a significant effect on relieve fatigue and sleep improvement.Above all, the study results powerful certified that: powder milk rich in opioidpeptides have a deeply effect on anti-fatigue and improving sleep.
