| We study the extraction of the polysaccharides of Auricularia auriculajudae in this paper.This paper confirms the extraction techniques of fruit body polysaccharides by orthogonal test,and the best medium and cultivation conditions of subsurface fermentation of agaricsmycelium. We also study the extraction, separation and purification of fruit bodypolysaccharides and agarics mycelium polysaccharides, and have a comparison betweenthem.The result is: the best extraction condition of agarics fruit body polysaccharides is115℃,90min, the ratio of agarics to water is1:35and the yield of crude polysaccharides is7.1gper100g. The optimization medium of subsurface fermentation consist of5g per100mLsucrose,0.5g per100mL yeast extract paste, the ratio of KH2PO4to MgSO4·7H2O is1:2(totalis0.3g per100mL). The cultivation conditions of subsurface fermentation of agaricsmycelium is pH5.42, temperature27℃, and tween8010g per100mL. The biomass ofmycelium and the concentration of polysaccharides are highest when the cultivation time is192h and216h respectively. We acquire the dynamics relations among the growth ofmycelium, the genesis of extracellular polysaccharide and the cruel sugar of substrateaccording to the cultivation conditions of subsurface fermentation of agarics mycelium.The result of paper chromatography component analysis is that agarics fruit bodypolysaccharides are divided into glucose and agarics mycelium polysaccharides are dividedinto mannose by sulphuric acid hydrolysis. The result of sephadex column chromatographicseparation show that the elution curve of agarics fruit body polysaccharides has3elutingpeaks which may explain it conclude three kinds polysaccharides,and the elution curve ofagarics mycelium polysaccharides saturated solution has two apparent eluting peaks. Theintrinsic viscosity of agarics fruit body polysaccharides is20.326。It is profited for the growth of agarics mycelium and genesis of extracellularpolysaccharide that the carbon source is sucrose. The organic nitrogen is more suited for thegrowth of agarics mycelium according to the screen test of nitrogen source. The quantity oftween80can effect the concentration of DO, and it can inhibit the growth of mycelium whenthe quantity of tween80is high. When the mycelium is on stationary phase, the ability of genesis of extracellular polysaccharide is degraded.There are great differences between agarics fruit body polysaccharides and agaricsmycelium polysaccharides according to the comparison of paper and column chromatography.The molecular weight of polysaccharides and the monosaccharide by hydrolysis are different.And the viscosity of agarics fruit body polysaccharides is larger than agarics myceliumpolysaccharides from sense organ. |
