This paper researches on three methods extracting the eggshell membrane’s keratin and its feature. The three methods are alkaline method(NaOH), oxidative method(H2O2) and enzymatic method(alkaline protease). This paper Starts from the reaction principle, extraction yield, formula weight, structure integrity, etc. and also inquire into hydrolytic process, extraction result and keratin’s surface feature.First, The egg shell membrane as raw material, uses alkaline method(NaOH), oxidative method(H2O2) and enzymatic method(alkaline protease) the best extraction method that had reported, to extract keratin.Second, studies Many aspects of the three methods of keratins which are extraction yield, reaction rate, formula weight, structure. The results are as follows:In the extraction yield aspect, oxidation (24.33%)> alkaline (19.31%)> enzymatic (15.19%); In the reaction rate aspect, the alkaline’s reaction rate is fastest, the enzyme’s reaction rate method is slowest; In the formula weight aspect, enzymatic (alkaline protease,31-43kDa)>alkaline (NaOH,14.4~20.1kDa)>oxidation (H2O2,<14.4kDa). The three methods’ formula weight’s distributions are all centralized, which shows their products’s quality is good. In the structure aspect, two extraction methods, that alkaline(NaOH) and enzymatic(alkaline protease) have preserved the keratin’s secondary structure integrity, oxidation(H2O2) has damaged keratin’s secondary structure.Third, The paper finally explores the use of three methods for extraction of keratin and the quality in the actual production. In the keratin molecular weight and structural integrity, superior oxidation enzymatic and alkali method, but in the small molecule polypeptide protein or amino acid products, we selected oxidation. If in the actual production requirements on time, we can use complex method, namely first with alkali method to loose keratin molecule structure, then with the reduction method or enzymatic method, this is a very good solution to the problem of long time alone to extract. Of course, in the actual production of small molecular keratin amino acids or peptides, we can first with alkali treatment, then with enzymatic method, so it is good to reduce the amount of enzyme.
