As an important antioxidant, glutathione has important physiological functions’such as removing free radical, detoxification and improve the effectiveness of the human immune system. In this paper, the extraction methods and extraction conditions of GSH in the yeast were studied by using wheaten food yeast as raw material and a preliminary study on the stability of GSH from yeast extract. Study on the Purification of GSH from yeast extract with D001×7(32) Macroporous Strongly Acidic Styrene Type Cation Exchange Resin. The main results were the following:The hot-water method, ethanol method and low temperature method were chosen to extract GSH from yeast by using extraction rate as index, it turned out that the best method is hot-water method. The best extraction conditions were determined preliminarily through single factor experiment which selects extraction time, extraction temperature, extraction pH and solid-liquid ratio as four factors. Extraction conditions were further optimized by using orthogonal experiment methods. The optimal extraction craft of GSH were:extraction time10min, extraction temperature90℃, extraction pH3, solid-liquid1:9. Under the optimal conditions, the extraction rate was89.40%.Test yeast extract by the TLC and HPLC method.The stability of GSH in extracting solution was studies by using retention rate as index. The time variation of GSH stability under different temperature and different pH value was investigated compared with the control which added L-Ascorbic Acid. The results showed that the stability of GSH was decreasing as the rising of temperature and pH value. Antioxidant Effect of L-Ascorbic Acid increased with the increase of temperature under a certain pH value. The better addition amount of L-Ascorbic Acid was0.5%, higher or lower amount of L-Ascorbic Acid can affect the stability of GSH.The antioxidation effect was better when keep pH4and the effect was bad in alkaline condition.Take adsorption rate as index, the optimal conditions of GSH purification by using D001×7(32) resin were determined:sample pH5, sample flow velocity1.OmL/min, elution concentration was0.5moL/L NaCl by using washout rate as index.GSH white lyophilized samples was gained finally, its purity increased1.07times. |
