Chitosan is a kind of unique natural polymer alkaline polysaccharide whichhas excellent biological compatibility, biodegradability, non-toxic pollution-freecharacteristics and other excellent properties, chitosan has been popularly used inmedicine sustained-controlled-release system. Montmorillonite (MMT), a kind oftypical layered silicate minerals, having good biocompatibility, non-toxic andside effects, has been widely used in food, medicine, paper, and chemicalindustries. Acetaminophen (paracetamol) is metabolite of Phenacetin in body,has the very strong antipyretic analgesic action, oral absorb quickly, in fluiddistribution in uniform, but short half-life typically14h, patients need to dailyuse of three to four times. If the ball is made slightly, can reduce the number ofmedicine, improve patient compliance. The microspheres were characterized byFourier transform infrared spectroscopy, Scanning electron microscopy and thetest of cross-linking degree.In this paper, ACAP-loaded chitosan microsphere were prepared withchitosan as carrier and glutaraldehyde as cross-linking agent. The preparation ofchitosan microspheres was optimized by orthogonal analysis, then the bettersynthetic technology condition was ultimately determined. The releasemicrospheres were characterized by Fourier transform infrared spectroscopy,Scanning electron microscopy and the test of cross-linking degree. The resultshowed that chitosan and glutaraldehyde had cross-linked. The cross-linkingdegree of microspheres was90.1%. By slow-release, we know thatacetaminophen of microspheres release completely in12hours.The montmorillonite was modified used the octadecyl quaternary ammoniumsalt and chitosan-montmorillonite composite materials were prepared. Using FT- IR and XRD characterized the two materials, we know that the layer spacing ofmodified montmorillonite had been obviously increased. Chitosan successfullyencapsulated in the layer of montmorillonite and the layer spacing ofmontmorillonite increase from1.23nm to2.46nm.The chitosan-montmorillonite-ACAP microspheres were successfullyprepared by emulsification-chemical cross-linking method. The preparation ofcomposite microspheres was optimized by orthogonal analysis, then the bettersynthetic technology condition was ultimately determined. Using FT-IRcharacterized microspheres structure, we know that ACAP successfullyencapsulated in composite microspheres. Compared with two release fertilizer,the drug release rate, the encapsulation efficiency and ACAP sustained releasetime of composite microspheres were obviously improved.
