The surface enhanced Raman spectroscopy has advantages of high sensitivity, range detection and fast in situ analysis, and it has been successfully used in the en vironmental science, medical, biological and other fields, people have pay more and more attention to the surface enhanced Raman spectroscopy (SERS).Although the SERS technique has many advantages, it is not popular to use this technique to detect the samples which are complex or trace concentrations directly. The interference of the complex samples of ingredients in the detection of actual sample SERS signal is the main reason about less application of SERS technique. To improve the SERS technique of selectivity and sensitivity are the key factors in the detection of trace substances in the complex sample. To change the actual sample and modify with nanoparticles are the dffective ways.The advantages of porous polymer capillary columns include diverse surface chemistry, high porosity, pH-stability, biocompatibility, low resistance to hydraulic flow, etcl. Porous polymer monolithic columns have been accepted as useful stationary phase for SPE and rapid liquid chromatography. The molecularly imprinted polymer monolith which combine molecularly imprint and monolith is a kind of p reparation sample methods potentially. But it is rarely reportd to use the monolith as SERS detect base. The polymer monolith modified with gold namoparticles thro ugh assembly in this research. This research expand the application of this function alization of monolith material and form the platform of preparation, separation, enr ichment and detection the sample. The main contents includes the following three parts:1:By investigating the influence on the polymerization reaction of the hole agents ratio, reaction temperature and crosslinking agent content, we prepare GMA polymer monolith and BMA polymer monolith in the best polymerization conditions.2:The different particle size of gold nanoparticles were synthesized use sodium citrate reduction method. Porous polymer monolith modified with gold nanoparticles were prepared by the method of chemical assem bly. We explored the applications of the polymer monolithic towards SERS by4-ATP molecles. We compare the influence of different particle size of nanometer gold modification to the SERS signal and obtain the best SERS enhancement effect when choose the diameter of50nm gold nanoparicles.3:By investigating the influence on the polymerization reaction of template and mono mer ratio, hole agents ratio, crosslinking agent content, we prepare BPA mol eculary imprinted polymer monolith in situ polymerization. The BPA molecularly imprinted polymer monolith modified with gold nanoparticles after the template was eluted. And it was used as a SERS substrate material for the detect of target molecules. The enrichment of molecularly imprint can be used for the detect of low content component, and it can improve the detection sensitivity.
