Chrysanthemum is a kind of asteraceae perennial herb that commonly used intraditional Chinese medicine and homology of medicine and food plant. The color ofchrysanthemum stem is light green or brown, basal half lignifications. Pickingchrysanthemums at the same time would produce a large number of stems, leaves andyield larger than the chrysanthemum. In order to make full use of the resources andincrease the added value.in this experiment, we used the chrysanthemum straw asmaterial,adopted water extraction research preparation process of low purity chlorogenicacid and made a simple process design, we hoped that thisresearch could provide someguidance and help for the industrialized produce of high purity chlorogenic acid.Themainresults wereshown as follows:In this study, we used water as solvent by cellulose-assisted extraction method toextraction the chlorogenic acid in chrysanthemum straw, and optimized the extractionprocess by single factor and orthogonal experiments, the results show that: the optimalparameters were soaking time30min,plus0.06%of the amount of enzyme,hydrolysistime60min, reaction temperature50℃,extraction temperature90℃, extraction time was45min, solid-liquid ratio is1:30, extraction times3times, among those factors, thesoaking time, extraction temperature and solid-liquid ratio have extremely effect on theextraction rate, however the extraction time has ordinary effect on the extractionrate.under the best Parameter extraction, we detected the water extract with HPLCmethodand reached a result that chlorogenicacid extractionratereached to96.83%.Used inorganic ceramic membrane technology and reverse osmosis membranetechnology to purify and concentrate The water extract.take The retention rate ofchlorogenicacid as the main indicators, referencemembraneflux,comparedand analysisprocessing effect of three different pore size ceramic membrane, the experiments showthat: the pore size of0.05μm has the best affection, made the chlorogenic acid retentionrate to85.93%, concentrated by reverse osmosis membrane, can removal74%of thewater of the water extract, the content of chlorogenicacid in concentrated liquid reachedto6.524mg/100mL.Utilized spray drying technology to make the concentrated liquid dried and toprepared dry powder in low purity of chlorogenic acid. optimized the drying effect by regulate the threemain factors of inlet air temperature, feed flow rateandatomizer speed,the experimentsshow that the best condition is: the inlet air temperature145℃, the feedflow rate20mL/min, the atomization speed26kr/min, and under the condition of airtemperature90℃, theeffectof thespray is preferred.Conducted the pilot scale test under optimized process:Invested1.05kgchrysanthemum straw,obtained22L concentrate and utilized spray dryingtechnology wegot89.92g dry powder products.thecontent of chlorogenicacidwas8.981%,higher thanthe content by smallscaleexperiment.By high performance liquid chromatography to detect the chlorogenic acid inconcentrated liquid and dry powder. The HPLC conditions: Agilent TC-C18,the mobilephase consisted of0.5%acetic acid (A90%) and acetonitrile(B10%),the flow rate was1.0mL/min, the columntemperature was35℃,the detection wavelength was325nm.theInjection volume:10μL. The linear equation was y=2.45E+04x-1.25E+05, r2=1, theminimumdetection was0.05μg/mL (S/N=5),the average recoverywas98.86%, RSD was0.8683%(n=6).The content of chlorogenicacid inChrysanthemumstraw was1.75mg/g.
