Objective:To investigate hydrolysis condition of plantain flavone glycoside,and estable a method to detectmine the content of flavonoid aglycones in plantain, optimize the extraction process of flavonoid aglycones. Then,study on the extraction technology of superfine grinding of plantain flavonoids, to explore extract flavones optimum technology of ultra fine powder in plantain.Medthods:In this dissertation, an HPLC analysis method to determine the content of total flavonoid aglycones in Plantago asiatica L. was established firstly. And quercetin, luteolin and baicalein were selected as reference substance. Then, the hydrolytic condition of flavonoid glycosides has been established. The best hydrolytic condition of flavonoid glycosides of weed of Plantago asiatica L. by reflux with orthogonal experiment was optimized. L9(34) orthogonal test has been used to evaluate the best hydrolytic technology of flavonoid glycosides which was evaluated by the content of total flavonoid aglycones including the concentration of hydrochloric acid, hydrolysis temperature and hydrolysis time. Finally, the best extraction of total flavonoids of weed of Plantago asiatica L. by superfine grinding with orthogonal experiment was optimized. L9(34) orthogonal test has been used to evaluate the best extraction technology of total flavonoids which was evaluated by the extraction efficiency of total flavonoids including ethanol concentration, material-liquid rate, extracting hour and extracting temperature.Results:The best hydrolytic condition was as follows:the total flavonoids of Plantago asiatica L. was extracted (50mg/mL) with5mol/L hydrochloric acid at75℃and hydrolytic time was1.5h. The best extraction technology was as follows:using60%ethanol as extractor with the ratio of material to liquid at1:25and extraction for15min at60℃. The experiment result showed that the contents of quercetin, luteolin and baicalein were1.36mg/g,0.35mg/g, and0.89mg/g, respectively;the content of total flavonoids was2.60mg/g, and the content of total flavonoids with ultra fine powder in plantain was3.35mg/g.Conclustion:This method is simple, efficient and accurate, which can be used for the quality control of total flavonoid aglycones in Plantago asiatica L.. The technology of superfine grinding can improve the plantain flavone aglycone extraction rate.
