| As the complexity and the number of samples increase, chromatographic researchers put forword higher requirements of separation efficiency by analytical instruments. Super Performance Liquid Chromatography (referred SPLC) which can achieve rapid and efficient separation has become an inevitable developing trend.In this paper, the SPLC system was constructed, on the basis of conventional HPLC system, which could achieve the goal of rapid and efficient separation. The performance of the system was evaluated then.The SPLC system successfully constructed could work under the pressure of60MPa. The relative standard deviations of some chromatographic parameters were less than3%, which could meet the requirements of qualitative and quantitative analysis by liquid chromatography. The SPLC system achieved the goal of efficient and rapid separation when combined with the use of sub-2μm columns, thereby reducing solvent consumption and saving the cost. Comparing with Waters UPLCTM H-Class system, the self-consturcted SPLC system owned considerable performance and lower cost, however, the system pressure and detective sensitivity was lower, which should be further improved.Then, chromatographic performances of the SPLC system were evaluated, including three small-particle C18columns and one sub-2μm column with s-trazine and amide embedded. The results indicated that three C18columns had their advantages, but their common disadvantages were that the peaks would tail severely when separating basic compounds. The sub-2μm stationary phase with s-trazine and amide embedded had better stereoselectivity so that it could shield the effect of residual silanols with analytes, which made up for the lack of C18stationary phase.By the evaluation of two types of reserved stationary phases on the SPLC system above, the SPLC system was proved to be applicable and provided a reference for the choose of columns in the separation of complex samples. |
