Surface enhanced Raman spectroscopy (SERS), as a kind of analysis and detection means, is widely used in studies of surface science, bolmedical detection, trace analysis, single molecule detectiondue to its advantages of high sensitivity, high selectivity and wide detection range. The substrates play a vital role in SERS and the research on the preparation of SERS active substrates remains a hot topic, its development will extend the application in other field.Monolithic column is a porous and uniform stationary phase by an in situ polumerization t in the capillary. It has the charictreistic of high specific surface area, good permeability, and the run-through porous structure. it is used in the modern industies for separation, purification, atalysis and so on. As an important kind of monolithic column, Inorganic silica monolithic column has abvious advantages in the mechanical strength, chemical stability compared to the other monolithic column. Moreover, for surface modification of chemical groups, silica monolithic column can supported avariety of stationary phase via a rich silanol groups on their surface and has the potential for separation of small molecules.In this thesis, the author use inorganic silica monolithic column modified with gold, silver colloid as SERS substrates, take the advantage of the monolithic column in separation and enrichment, achieved the detecting Raman spectra detection of trace substance in situ, Meanwhile, a reusable SERS-active substrate was prepared.The major contributions of this work are as followings:1. Inorganic silica monolithic column with dual-pore structure were prepared by sol-gel method. The effects of synthesis methods and conmpents, such as inner surface treatment, choice of precursor, PEG content, on the preparation of inorganic silica monolithic column were investigated. After Activating the monolithic column and bonded3-mercapto propyl trimethoxy silane to its surface, gold nanoparticles were immobilized on the monolithic column by the thiol. The results indicatd that a very good sensitivity compared with the nanoparticles unassembles by using p-aminothiophenol as probe molecules.2. Au nanoparticles were synthesized by citrate reduction method, and Sodium borohydride reduction method for the preparation of Ag nanoparticles. We investiaged the absorption of the nanoparticles on the surface of monolithic porous materials, and characterized it by UV-Vis spectroscopy, TEM and SEM.3. We developed a new method for preparing silica monolithic column by using CTMAB as supramolecular template and add APTES as a reactant at the same time, which can solve the problem of complexity and long preparing time. We also verified the limit and repeatability of detection, elution ability of silica monolithic column in SERS by using PATP、R6G、crystal violet as probe molecules.
