Biodegradation and transformation of pollutants is accomplished by enzymecatalysis in biological wastewater treatment, study biological wastewatertreatment system in microbial activity and its role in the process is of greatsignificance for in-depth to explore the mechanism of biochemical reactionsystem to improve the efficiency of sewage treatment. Catalase is anoxidoreductase capable of catalyzing the oxidation-reduction reaction in thebiological wastewater treatment process,the role of activated sludge catalasedecomposition of hydrogen peroxide can be toxic to microorganisms, to ensurethe efficiency of wastewater treatment. In order to in-depth research and analysisof the activated sludge system catalase activity characteristics and relatedfactors, to establish contact catalase and biological wastewater treatmentsystem,making it one of the indicators that characterize the activity of activatedsludge characteristics, further detecting catalase indicators to evaluate theperformance of the of sewage biological sewage system,guide and optimizewastewater biological wastewater system operation controlg,actual activatedsludge as an object,a series of experiments to study the catalase activitydetection method and impact factors.Activated sludge catalase (CAT) activity orthogonal test,it is found thatwhen the sludge volume to1.5ml,the amount of sulfuric acid was added to1ml, the amount of alum added to0.7ml, shaking time was20min, thedetected activitysludge catalase highest indicators. Under these conditions,anumber of test research on biological wastewater treatment system, CATactivity, COD, TN, NH3-N, TP removal variation, the results showedthat:the activated sludge CAT activity COD,TN,NH3-N removal of a high degree of correlation, system in COD, TN, NH3-N removal is relativelyhigh,but not sensitive to TP removal.Experimental study of the biological wastewater treatment system,pH,temperature,MLSS,C/N ratio of the metal ion of sludge CAT activity,theresults showed that,CAT activity is very sensitive to the pH value,CATactivity is very low in acid and alkali state,higher activity in the neutral weakbase state,corresponding to the optimum pH value of7;temperature is too lowor too high,inhibition of CAT activity,especially when the temperature ishigh,the impact of thermal denaturation than the warming caused by reactionacceleration greater impact, excessive temperature even cause catalaseinactivation,catalase optimum temperature was25℃;CAT activity increasedwith the MLSS concentration increased,CAT activity MLSS concentration of6000mg/L;with the improvement of the C/N ratio,CAT activity increasedslightly,but the increase is not obvious,catalase optimum operation of C/Nratio of10;have a greater impact on the CAT activity of activated sludgebiological wastewater treatment system containing metal ions,several metalions on the CAT activity impact is not the same,Mn2+can be increased CATactivity of the activated sludge,Mg2+and K+weaker activation,Zn2+,Li+,Ba2+and Ag+CAT activity has a certain inhibition,and affect the removalefficiency of the hydrogen peroxide in the sewage,A13+exists,make catalaseinactivation.Construction of the mathematical model between catalase experimentalfactors and catalase activity,seen by the model, the experimental design withinthe level,acetate buffer pH value and CAT activity was a parabola openingdown,that the pH value is too high or too low on its CAT activity have a certainextent;CAT activity and the temperature was down parabolic relationship,low temperature and CAT activity was a linear correlation relationship, as thetemperature rises into a parabolic relationship, lower temperatures withincreasing temperature to enhance the activity of CAT,CAT activity began todecline when the temperature is raised to a certain extent,this protein andenzyme characteristics consistent. MLSS and CAT activity was positivelycorrelated,with the level of MLSS value increased,CAT activity was slowafter the fast rise. In comprehensive consideration of the pH,temperature,MLSS,and C/N of the system of CAT activity by the model,C/N performanceof CAT activity is not very significant,the temperature and pH affect theactivity of the key factors of CAT activity;consider the combined effect ofvarious factors interact,pH7,temperature of25℃,the MLSS to8000mg/L,the highest value of CAT activity detected.
