Reverse micelle extraction is a new type of high efficient separation technology whichcan separate plant protein and oil at the same time, and peanut protein is of high nutritionalvalue. Therefore, it is of great significance to study the production and application of reversemicelle extraction of peanut protein. This paper mainly research and contrast the optimalprocess conditions of SDS and DTAC reverse micelle systems forward and backwardextraction peanut protein under the presence or absence of ultrasound effect. The optimizationprocess conditions of the forward and backward extraction using AOT reverse micelle systemextraction peanut protein are also studied. To explore the mechanism of the AOT reversemicelle system extraction of peanut protein, the system characteristics and medium scalestructure are studied. At last, the differences of the functional characteristic and the structureof peanut protein which extraction of different reverse micelle systems and extraction waysare studied and compared with peanut protein which extracted by alkali-solution andacid-isolation and that of drum drying method selling on the market.Using SDS and DTAC reverse micelle systems to extract peanut protein, the optimalprocess conditions of forward and backward extraction under the presence or absence ofultrasound effect are studied. The results show that using ultrasound-assisted reverse micellesto extract peanut protein, the forward extraction efficiency improves15%, and the backwardextraction efficiency improves10%, approximately, and also greatly shorten the extractiontime. The conditions of forward and backward extraction peanut protein using AOT reversemicelle system assisted by ultrasonic are researched. Using the orthogonal experiment designsto optimize the forward and backward extraction process conditions, and compared with theoptimal process conditions of SDS and DTAC reverse micelle systems. The AOT reversemicelle is the most suitable for extraction of peanut protein, and the extraction rate is higher.With the principle of dynamic light scattering, the medium scale structure of AOTreverse micelle is measured using a Zeta Plus instrument. The particle size distribution ofmicelles in the AOT reverse micelle solution of different concentrations and the particle sizedistribution of the particles in the corresponding anterior extract liquid are studied, then therelationships of particle size distribution and peanut protein extraction efficiency are included:when the particle size is mainly distributed in6.0nm~7.0nm, the extraction efficiency of peanut protein is the highest, and the AOT concentration is0.06g/mL right now.Compared with peanut protein of alkali-solution and acid-isolation extraction and that ofdrum drying method, the functional characteristic and the structure of peanut protein whichextraction of different reverse micelle systems and extraction ways are studied. It is found thatthe color, the solubility, water holding capacity, emulsifying and emulsion stability of peanutprotein extraction using reverse micelles are all better than the two others; the size of peanutprotein prepared by the reverse micelles is smaller through contrast the scanning electronmicroscope pictures of different proteins; The FTIR and DSC results show that the proteinsecondary structure of β-sheet and disordered structure turn into β-turn using reverse micellesextraction, but the protein don’t degenerate greatly; The study results of X-diffraction showthat using reverse micelles extraction protein can affect the microscopic crystal structure ofpeanut protein, but not significantly.
