The first part of this paper relates to the research on the separation method of highly polar compound DMC and impurities in it.The problem of separating and determining highly polar compounds is a hot research topic as well as a research challenge in the scientific world, which refers to proteomics, metabonomics, traditional Chinese Medicine analysis, natural product chemistry, synthetic drugs fields and so on. Methacryloyloxyethyl trimethyl ammonium chloride (DMC) is an excellent cationoic reagent. Industrial products of DMC usually contain some highly polar compounds, such as dimethylaminoethyl methacrylate (DM), methacrylic acid (MAA), dimethylaminoethanol (DMAE), choline chloride (CC), some oligomers and so on, which are very important factors influencing the quality and yield of the products. It is highly necessary to separate among DMC and impurities in it. We attempted some separation and analysis methods such as Reversed Phase High Performance Liquid Chromatography (RPLC), Hydrophilic Interaction Liquid Chromatography (HILIC), Ion Chromatography (IC), Liquid Chromatography and Mass Spectroscopy (LC-MS), Proton Nuclear Magnetic Resonance Spectroscopy (NMR), and so on. Finally we choosed to use RPLC and HILIC methods to separate and determine DMC and highly polar impurities may contain in it. It provides a reference for the problems of separating highly polar compounds which may often meet with in organic chemistry.The second part of this paper relates to the synthesis of derivatives of the lignans otobain.Natural medicine usually means active composition derives from all living things in nature, which is an important component of clinical medication. Lignans and its derivatives have the significant function of anticancer and antineoplastic, which arouse wide concern by scientists in the research of natural products. Hydroxyotobain was one kind of lignans extracted from the bark of Myristica Fragrans Houtt by Zhang Mei in our group. Using Hydroxyotobain as the lead compound, four terminal’ compounds had been prepared by Xu Zijin in our group, which were Didehydro-otobain (otobaene), Methoxyotobain, Allyloxyotobain and Benzyloxyotobain. It was proven that these compounds all have some anticancer activities. Zhou Bin in our group designed two Hydroxyotobain derivatives, and he synthesized the key intermediate of the two derivatives. On these bases, for the purpose of further research between the structure and bioactivity, we designed two another otobain derivatives. We made effects to the totally chemical synthesis of them. Finally, we got the objective products of the two otobain derivatives by bromination, carbonyl protection, nucleophilic addition, oxidation, michael initiated ring closure and so on, which originated from the simple compound piperonal.
