In recent years, in order to further explore and recognize the relationship betweenthe structure and function of DNA, people study a large number of interactions betweenmetal complexes and DNA. Explore the interactions between metal complexes andDNA mechanism, has become one of the hot research topics in the field of biology andchemistry. The interactions of DNA and metal complexes mainly include three types:intercalation binding, electrostatic binding, and groove binding. Intercalation isconsidered as one of the most important among the three kind, because of its providinguseful theoretical basis for the design and synthesis of DNA sensors and probes withapplication value. In this paper, we selected2,5-dimercapto-1,3,4-thiadiazole (DMTD),1,10-phenanthroline (1,10-phen), imidazo [4,5-f][1,10] phenanthroline (IP), dipyrido[3,2,a:2′,3′-c] phenazine(DPPZ) and2-(4-hydroxyl)-imidazo [4,5-f][1,10]phenanthroline(HOIP) as ligands, Mn(Ⅱ), Ni(Ⅱ), Zn(Ⅱ) as central ions to synthetizemetal complexes. The interactions between metal complexes and DNA were measuredby UV, fluorescence spectroscopy, electrochemical method and viscosity method. Andtheir binding ratio and binding constant were calculated.First, organic complex2,5-dimercapto-1,3,4-thiadiazole was synthesized accordingto the literature and characterized by infrared spectroscopy,[Mn(DMTD)(1,10-phen)2Cl](1) and [Ni(DMTD)2(1,10-Phen)2](2) were synthesized, and their structures weredetermined with X-Ray diffraction, elemental analysis, infrared spectroscopy, powderdiffraction. The interactions between (1) and (2) with DNA were measured by UV,fluorescence spectroscopy, electrochemical method and viscosity method, the resultsshow that the modes between them are intercalation.Second, imidazo [4,5-f][1,10] phenanthroline was synthesized according to theliterature and the structure was characterized using infrared spectroscopy and1H-NMR.The complex [Zn(DMTD)(IP)2H2O](3) which contains DMTD and IP has beensynthesized. The results of interactions between (3) and DNA show that the modebetween (3) and DNA is electrostatic interaction.Third, dipyrido [3,2,a:2′,3′-c] phenazine was synthesized according to the literatureand characterized by infrared spectroscopy and1H-NMR and complex[Zn2(DMTD)2(DPPZ)2](4) was synthesized. The results of interactions between (4) andDNA show that the mode between (4) and DNA is intercalation. Fourth,2-(4-hydroxyl)-imidazo [4,5-f][1,10] phenanthroline was synthesizedaccording to the literature and its structure was characterized using infraredspectroscopy and1H-NMR, complex [Zn(DMTD)(HOIP)2]Cl·H2O(5) was synthesized.The results of interactions between (5) and DNA show that the mode between (5) andDNA is intercalation.
