γ-polyglutamic acid (Polyglutamic acid, γ-PGA, γ-PGA) is formed of a combination of the Friends of the D-or L-glutamic acid, a-amino and γ-carboxyl group of the γ-glutamyl acyloxy anion key polymerizationthereof.γ-PGA is adhesiveness, non-toxic, edible, easy-to-film formation, flocculation, moisturizing properties and characteristics, it can be widely used in various fields, in the pharmaceutical industry as a drug carrier, as a thickener in the food industry, cosmetics the industry as a humectant, and it can also be used as a retaining agent in agriculture and fruit, vegetables antifreeze, a preservative used in agriculture, used as water treatment flocculant and a heavy metal sequestrant [1], it can also be a new kind of green plastic.γ-PGA is expensive, it can be widely used, and a huge demand for space, but now it is far failed to meet the needs of the market in the production and supply. Therefore,we constantly improve the inadequacies of the current study, to obtain a process route of γ-PGA production, it’s low cost production, easy to be producted.This project uses liquid fermentation synthesis of γ-polyglutamic acid, taking advantage of Bacillus subtilis batch fermentation, at first we separated to obtain a solution containing the product, and then precipitated γ-PGA make use of an organic solvent precipitation method combined isoelectric point, the extract solution was purified to thereby obtain a high-purity γ-PGA. molecular weight determination and nature detecting is the last procedure.In this project, we obtained the product--γ-PGA through employed the preferred strains fed-batch, in the meantime, achieved the automatic control of the fermentation process.The process route is simple, short production cycle, high recovery, no environmental pollution, green biosynthesis in the true sense. Final process route is exceed of national production standards.It’s not demanding requirements of the equipment and the experimental conditions in the fermentation and extraction and purification process.Aspartate Bacillus fermentative production of γ-PGA reached more than30g/L, and by optimizing the fermentation process and process the end of the experimental data:10L fermenter, γ-PGA production≥70g/L glutamate≥80%conversion rate.
