For the problem of the outbreak and the effective use of the large area of sea grass inQingdao coastal, the author made a lot of experimental study and optimized the method forextracting the monosaccharide, and made the extraction rate improve greatly. A method for theanalysis of monosaccharide composition and its content in enteromorpha’s polysaccharide wasalso established in this study. And the author used the method to analyze several importantcomponents as well as the safety parameters of enteromorpha linza in Qingdao coastal, and alsotried to analyze its nutrition and safety, provided a basis technical support for the followingstudies.The extraction rate of polysaccharide increased by more than15%by simulating thesaccharification of starch during the wort manufacturing operation, adopting complex enzymeand temperature programming to extract the polysaccharide. The study found thatphenol-sulfuric acid method which is widely used currently to determine the algaepolysaccharide is unreasonable. It is unable to analyze the composition of the polysaccharides.The experiment established the pre-column derivatization high performance liquidchromatography method to determine the enteromorpha’s polysaccharide by optimizing thecondition of pretreatment, This method increases the extraction rate by more than15%. Inaddition, condition optimization still included the chromatographic conditions and thederivatization conditions, especially the optimization of mobile phase. The external standardmethod was used for the quantitative analysis in this method. Under the optimized experimentalconditions,8monosaccharides were separated completely within65minutes. The calibrationcurves were linear in the range of1-100μg/mL for8monosaccharides with correlationcoefficients over0.999. The average recoveries for8monosaccharides were between76%and108%, with relative standard deviations (RSDs) less than10%. The rhamnose was found to bethe main component in enteromorpha’s polysaccharide by this method. Glucuronic acid, glucose,xylose, galactose, a small amount of mannose, galacturonic acid and fucose were also found inenteromrpha’s polysaccharides. There was a significant correlation between the main functionalgroup sulfate and rhamnose(R=0.928), so the rhamnose was suggested to be the main index tomeasure the enteromorpha’s polysaccharide and enteromorpha powder. In addition, the structureof the polysaccharide was complex, and the sum of monosaccharide can not accurately expressthe content of enteromorpha’s polysaccharide, but at present, we can use the sum ofmonosaccharide and the content of rhamnose to characterize the polysaccharide levels, whichwas similar to the Phenol-Sulphate method.By horizontal and longitudinal comparison of enteromorpha linza in Qingdao andenteromorpha in other waters as well as some other common algaes, it has been found that thereare abundant kinds of polysaccharide with high content in enteromorpha linza in Qingdao.What’s more, comparing with the other enteromorphas and common edible algas (such as kelpand nori), the content of uronic groups and sulfate groups in enteromorpha linza are high, and the value of its polysaccharide is great. Through the detection of the relevant safety parameters andpreliminary evaluation of the other nutitional indicators, it displayed that enteromorpha liza inQingdao is a kind of alga, which is full of protein, aminoacids, fatty acids and mineral substances,and its fat is low. Enteromorpha liza in Qingdao has a comprehensive nutritional value and it issafe to eat.The research was conducted to promote the edible value of enteromorpha linza in Qingdaocoastal, and made it had a large-scale exploitation and utilization, and prevented the waste ofresources during the outbreak. The results of this study provided a basis for future research of theevaluation criteria for enteromorpha’s polysaccharide and enteromorpha powder. Now it isurgent to establish the quality standard to regulate the functional food and raw material, andcontinuously improve the quality of functional food and promote the healthy and rapiddevelopment of the enteromorpha products.
