The pigskin scrap as raw material, the use of a meat grinder for collagen polypeptide collagen the pigskin scrap crushed into pigskin debris, alcalase and papain hydrolysis pigskin debris.Degree of hydrolysis by the protein as an indicator, explore the impact of temperature, hydrolysis time, substrate concentration, pH value, enzyme dosage on Alcalase production pigskin collagen polypeptide, found that temperature, hydrolysis time, pH value, the amount of enzyme affect the degree of hydrolysis of pigskin collagen significantly. By response surface methodology to determine the optimum conditions temperature of54.98℃, enzymatic time5.01h, substrate concentration5%, pH8.50,the amount of enzyme3223.94U/g substrate, pigskin collagen under optimal conditions, the degree of hydrolysis of13.63%;Protein degree of hydrolysis of the indicators, examined the temperature, hydrolysis time, substrate concentration, pH value, the amount of the enzyme on papain production pigskin collagen polypeptide, the results show that the temperature, hydrolysis time, pH value, the amount of enzyme impact on the degree of hydrolysis significantly. Using response surface methodology to determine the optimum conditions for use of the the papain preparation of pigskin collagen polypeptide: temperature58.54℃, enzymatic time4.79h, substrate concentration of5%, pH6.70the enzyme dosage2394.98U/g substrate under the conditions of a degree of hydrolysis of16.53%; The use order of Alcalase and papain little effect on the degree of hydrolysis of the pigskin collagen.Pigskin collagen protein hydrolyzate peptide for the study, and the study of the nature of the polypeptide. The study found that the the this experiment pigskin collagen polypeptide absorbent0.17±0.02g water/g sample, an oil absorption of1.27±0.03mL soybean oil/g sample; Pigskin collagen polypeptide has good water-holding capacity, although not as well the glycerin water holding capacity, but the water-holding capacity was significantly better than water;30g/L pigskin collagen peptide emulsifying activity to47.9±0.5%, stored at25℃37±0.5%after5h emulsion stability; foaming149.7±0.5%, stored60min foam stability5.2±0.4mL;32mg/mL the pigskin collagen polypeptide clearance rate of hydroxyl radical (·OH) was99.51%;50mg/mL the pigskin collagen polypeptides has DPPH· radical scavenging rate of83.48%, a clear rate of79.42%on the superoxide anion radical (O2-·); The pigskin collagen polypeptide on the reducing power of the iron ions was less strong than VC, but a certain reduction ability, and was capable of chelating ferrous ions to a certain extent.Using SDS-PAGE analysised pigskin the molecular weight distribution range of the system pigskin collagen polypeptide, and found that the the pigskin after the Alcalase and papain obtained collagen polypeptide has a molecular weight of smaller, and the molecular weight distribution was concentrated. Polypeptide molecules obtained in this test was mainly concentrated in20.1KD-14.3KD, many polypeptide of molecular weight less than14.3KD.As raw material, fat in the pigskin scrap obtained lard, and study on lard refining process. Opted oil4%by weight of hot water, then add0.15%salt, slowly stirring, hydration3times for optimum hydration conditions; Selectlye concentration of5%, the ultra amount of25%of the theoretical amount of alkali, alkali the alkali refining the initial temperature of50℃,alkali refining the final temperature of75℃for the alkali refining the optimum conditions; The best decolorization conditions:bleaching adsorbent dosage of2%, the operating temperature of100℃, the operating time of15min; the best deodorizer conditions:deodorization pressure of0.1MPa and a temperature of120℃, the deodorization time of15min.
