| Sargassum Kjellmanianum is a perennial macroalgae which blongs to the familyof Sargassaceae, and the Genus of Sargassum. It mainly distributed in the cold waterof the east coast of China. Due to its special environment of growth, some products asproteins, polysaccharides which has special biological activity are synthetized. Thesesubstances provide vast resources in search of natural medicine. And the activesubstances from seaweeds has become hot spots in research of natural medicine.Fucoidan is a kind of sulfated polysaccharides prevalent in brown algae, and manystudies have shown that fucoidan has several biological activities such as antioxidant,immune regulation, proliferation inhibition of tumor cells. Besides, its curative effectin the treatment of chronic renal failure has also been confirmed in recent years.However, due to its complexity in structure and compose, its mechanism of biologicalfunction has not been clearly understood. The raw material of fucoidan are limited tothe Laminaria, Sargassum fusiforme, and the polysaccharides from other alginatesuch as Sargassum Kjellmanianum has been studied less comparatively. In this paper,the sulfated polysaccharide was prepared from Sargassum Kjellmanianum which werecollected in Shandong coastal. Different sulfate content fucoidans were preparedduring ultrasonic assisted radical degradation, chromatographic separation techniquesand sulfated modification. On the basic studied of the physical and chemicalproperties and structure analysis, we studied its antioxidant activity, anti-tumoractivity in vitro and in vivo, immunomodulatory and moisturizing activity.(1) SKP was extracted from Sargassum Kjellmanianum using the hot waterextraction method. SOP was prepared during sulfated modification to SKP.SKP1,SKP2and SKP3were obtained after ultrasonic assisted radical degradation andgrading by DEAE-52cellulose column chromatography. The sulfate content of SKP,SOP, SKP1, SKP2and SKP3were18.9%,36.7%,8.4%,25.6%and29.9%,respectively.(2) The chemical composition analysis revealed that the five fucoidancomponents were mainly composed of fucose, galactose and mannose, followed by a small amount of xylose, glucose, arabinose and rhamnose, and the composition aredifferent. The content of uronic acid of SKP was23.49%which redused afterdegradation and grading. While total sugar content was increased after grading.Infrared spectroscopy results show that the five fucoidan fractions showed thecharacteristic absorption of the sulfate group.(3) In vitro and in vivo immune and antitumor was studied, and the results showthat three fucoidan fraction promote the proliferation of immune cells. Thereinto,SKP2and SKP3showed significant effect on spleen lymphocyte promotion andmacrophage proliferation which can also enhance macrophage phagocytic ability ofneutral red and the release of NO. In vitro and in vivo antitumor results show that,fucoidan fractions had certain inbibitional effect on Hela cells, and had little on HT-29in vitro. SKP2and SKP3significantly inhibited the proliferation and spread of theS-180tumor in mice, and presented dose-dependent manner.(4) Fucoidan fractions had significant antioxidant effect which mainlymanifested in the superoxide anion radical and hydroxyl radical scavenging effect.SKP3had the most obvious effect on·OH and O2-·scavenging, and the clearance rateof it can up to79.09%and68.22%. The scavenging effect on the free radical DPPHwas also significant, and the clearance was dose dependent while increased with theimprovement of the sulfate content.(5) Results of moisturize and moisture absorption study show that SKP has goodmoisturizing and moisture activity and its moisturizing and moisture absorption ratecan up to67.89%and49.22%respectively. The moisture absorption ratecorresponded to HA, and moisture activity was a little better than glycerol.Moisturizing and moisture activity of fucoidan fractions after degradation weredecreased which had little differents among the three fractions SKP1, SKP2andSKP3. |
