The natural pigment from microbe had antibiotic activity and pha-rmacological functions. Using microbe to produce natural pigment w-as not only to overcome several shortcomings produced by animals and plants, but also liable to industrialization.The silica gel column was used as purification method to obtain pure sample. The sample was shown to be prodigiosins by HPLC a-nd LC-MS. The compounds was shown by mass and nuclear magne-etic resonance spectroscopies to be two known prodigiosin analogs: metacycloprodigiosin and undecylprodigiosin.We investigated the effects of two prodigiosins against human he-patoma cell line(HepG2), human colcnic carcinoma cell line(HCT-116) and human gastric carcinoma cell line(SGC-7901) using MTT assay. The undecylprodigiosin exhibited potent in vitro activity against the three cancer cell lines(IC50:14.8,50.4,20.3μM/L) compared with metacycloprodigiosin.The optimization of culture medium was carried out with Respon-se Surface Methodology(RSM). The result showed that the optimum concentration of carbon, nitrogrn, microelement and amino acid was76.82g/L,20.54g/L,21.38pmol/L,22.96mmol/L. In the meantime, The best fermentation condition:inoculum3%, liquid volum40mL, pH7.0, shaking speed200r/min, temperature28℃was got by ort-hogonal test. The yield of undecylprodigiosin was increased86.25%compared with initial strain.
