The active ingredients of Dioscorea opposita Thunb and Rehmannia glutinosa which arefamous geoherbs in Henan province were reaserched. The studies focused on the influence of thecomponent and content caused by the compatibility of Dioscorea opposita Thunb and Rehmanniaglutinosa by forming Medicine Pair. The main contents are as follows:1. The research of Medicine Pair, the chemical components of Dioscorea opposita Thunb andRehmannia glutinosa and the methods for the determination of the content of their main activecomponents in recent years were briefly reviewed. The research subject was put forward on thisbasis.2. The methods for the determination of the content of their main effective components in theMedicine Pair of Dioscorea opposita Thunb and Rehmannia glutinosa were developed. The highperformance liquid chromatography (HPLC) method for the simultaneousdetermination ofcatalpol,5-hydroxymethyl-2-furaldehyde (5-HMF) and adenosine was developed. The HPLCmethod was developed to separate several monosaccharides efficiently by precolumnderivatization with1-phenyl-3-methyl-5-pyranozolone (PMP). And the method was successfullyused for the first time to determine and compare the monosaccharide composition and content ofpolysaccharides in the Medicine Pair of Dioscorea opposita Thunb and Rehmannia glutinosa andtheir single herb medicine. The quantitative determination of polysaccharides content in theMedicine Pair of Dioscorea opposita Thunb and Rehmannia glutinosa was conducted by usingphenol-sulfuric acid. And the polyphenolics content in the Medicine Pair was assayed by usingFolin-Ciocalteu.3. The developed HPLC method for the simultaneousdetermination of catalpol,5-HMF andadenosine was applied to determine the contents in the Medicine Pair of Dioscorea oppositaThunb and Rehmannia glutinosa with different proportions (3:1,2:1,1:1,1:2,1:3,1:5). Theresults showed that the compatibilities of the Medicine Pair composed by Dioscorea oppositaThunb and Rehmannia glutinosa could promote the dissolution rate of catalpol. But thecompatibility of the Medicine Pair composed by Dioscorea opposita Thunb and Radix RehmanniaPreparata had inhibitory action to the dissolution rate of5-HMF. The content of adenosine in theMedicine Pair had no obvious change with the variation of the proportions of Medicine Pair.4. The monosaccharide composition and content of polysaccharides in the Medicine Pair ofDioscorea opposita Thunb and Rehmannia glutinosa and their single herb medicine weredetermined and compared by the establed HPLC method. The results showed that all threepolysaccharides contained mannose, rhamnose, glucuronide acid, glucose, galactose and arabinose,however, fucose was not detected. At the same time, it was found that the content of eachmonosaccharide in the Medicine Pair was in the content range between the single Dioscoreaopposita Thunb polysaccharides and Rehmannia glutinosa polysaccharides. 5. The determination of the contents of polysaccharides and polyphenolics in the MedicinePair of Dioscorea opposita Thunb and Rehmannia glutinosa and their single herb medicine wereconducted by ultraviolet and visible spectrophotometry. The results showed that thepolysaccharides content in Dioscorea opposita Thunb, Rehmannia glutinosa and their MedicinePair (1:1) was3.50%,6.63%and8.60%, respectively. The polyphenolics content in Dioscoreaopposita Thunb, Rehmannia glutinosa and their Medicine Pair (1:1) was0.076%,0.263%and0.192%, respectively.
