| We chose the Fermented grain samples, Daqu and pit mud samples as our Study object using PCR-SSCP method, and preliminarily analyzed the changes of pronucleus microorganism’s community in which provides theoretieal basis for development of liquor. The results are as follows:1. Optimization of the methods and data of PCR-SSCP. First, we found out the suitable DNA extraction method each for fermented grains, Daqu and pit mud samples. Second, found a best silver staining method by compared different methods. Third, we found out the suitable PCR-SSCP conditions. Research show that:volume ratio between denaturing buffer and sample were2:1, the gel concentration was14%, acrylamide and bisacrylamide concentration was49:1, reaction conditions under pressure of300V,18h,4℃.2. The pronucleus microorganism’s community were similar and also had the polymorphism in each different fermented grain samples. The results were as follows from SSCP map:bands from No.12to No.22were showed clearly in all fermented grain samples. But the changing laws of dominant degree among bands were different. Besides, the mean diversity index of all samples kept between2.40and2.94. The similarity index of PCR-SSCP pattern of samples from different spots in the same pit was higher.3. The results showed:The pronucleus microorganism’s community was similar and also had the polymorphism in each different samples.The diversity indexes of different stage of Daqu microorganisms were change a lot in each simple of various fermentation stage, different micro flora had complex ecology effects of coordination and the restriction.4. Bands from No.7to No.16were showed clearly in all pit mud samples. The pronucleus microorganism’s community of the same pit during different fermentation period were similar. Besides, the mean diversity index of all samples kept between1.95and2.83. With the increasing of pit age, the diversity index of samples from the same spots generally showed increasing trend. The similarity index of PCR-SSCP pattern of samples from different spots in the same pit was higher, between0.92and0.98. While the similarity index of samples from different pit ages was lower, between0.78and0.96. |
