The Study On Anti-Obesity Efficacy Of Polyphenols Of Ilex Kudingcha C.J.Tseng

With the improving of people’s life, the incidence of disease of obesity increase everyday. Preventing and curing these kinds of disease have been the foucus of global healthy in dustry. Searching natural, high effect safe medicine and healthy food for diposity have been the roject that global seience and technology field and product field care mostly.Kudingcha is a kind of tea drank by people from north and east part of China. Ilex kudingcha C.J. Tseng is a one of the main breed varieties of kudingcha and its active ingredients as well as healthy protection functions have been more and more concerned in recent years.Polyphenols extracted from kudingcha is an activated substance with special physiological functions,such as reducing blood fat,anti-oxidization,resistance to free-radical etc. This project aims at isolation, purification of Ilex kudingcha C.J.Tseng polyphenols, evaluation the function of Ilex kudingcha C.J. Tseng polyphenols, such as anti-obesity the inhibitory effects of polyphenols on a-amylase and prancreatic lipase.HP20was selected as the medium for initial isolation of polyphenols.The method to initially separate polyphenols was established using column chromatography.Total polyphenols(TP)was separated into two fractions which were named TP1,TP2after gradient elution with water and70%ethonal.Then, the TP2was separated into three fractions which were3,4-diCQA,3,5-diCQA,4,5-diCQA with30%methanol(contain1%formic acid)by BUCHI colomn.The anti-obesity activity of polyphenols(TP、TP1、TP2) extracted from Ilex kudingcha C.J.Tseng was evaluated through its inhibitory ability towards a-amylase and pancreatic lipase.The result showed that kudingcha polyphenols can strongly inhibit activity of a-amylase and pancreatic lipase. And the three parts showed non-competitive reversible inhibition effects on a-Amylase.The inhibitory ability of a-amylase and pancreatic lipase is:TP>TP1>TP2.In accordance with the rat model for obesity prevention, based on the observed indicators,results showed that: With obesity compared with model group, the polyphenols can reduce the increase of the weight of rats and reduce the Lee’s index, liver weight, epididymal fat factor. The polyphenols from Ilex kudingcha can also low the serum and liver homofenate level of TC,TG,LDL-C,MDA and the activities of LPS,increasing HDL-C level and the activities of SOD,GSH-PX. The results showed that the Ilex kudingcha has anti-obsity effect.
