Maize pollen, which is especially rich in the variety of anti-radiational factors,is a very important resource of Medicinal and Edible food. In this paper, maizepollen was picked out as the research material. Maize pollen polysaccharides,flavonoids and water extracts were extracted by different separation methods, notedas PMP, FMP and TEMP respectively. Anti-radiation effects of PMP, FMP andTEMP were verified and compared in vitro and vivo experimental studies. Further,we analysed the composition of active flavonoids. The main conclusions of thisstudy are listed below.Maize pollen polysaccharides were extracted in the hot-water system andprecipitated with athanol of different strengths in turn. Flavonoids were extracted bya ultrasonic method and purification by AB-8macroporous resin. The water extractswere extracted by hot-water with ultrasonic processing. The extraction rate ofPMP, FMP and TEMP were1.14%,0.075%and47.3%respectively.The antioxidant activitives of PMP, FMP and TEMP in vitro were conducted bydetermining the reducing power, hydroxyl radical scavenging activity and2,20-azino-bis-(3-ethylbenzthiazoline-6-sulfonic acid)(ABTS) radical scavengingactivity. According to comparison and analysis of experimental resultscomprehensive, the maize pollen flavonoids fraction (FMP) is deemed to havestronger antioxidant activity in vitro.The Antioxidant enzymatic activities and immune system reaction of mice,administered intragastrically with PMP, FMP and TEMP and induced by3Gy γ-rayin turn, were determined. The liver and spleen index (liver or spleen weight/bodyweight1000), the counts of peripheral white blood cells (WBC) and theendogenous antioxidant enzymes (GSH-Px and SOD) activities were elevated. Themaize pollen fractions also can decrease the organism’s MDA levels and inhibit thereaction of lipid peroxidation in organism simultaneously. According to theexperimental results, the FMP is deemed to have stronger anti-radiation activity invivo.The protein composition in the mice spleen were demonstrated bytwo-dimensional gel electrophoresis (2-DE) and identifed by liquid chromatographytandem mass spectrometry analysis (LC-ESI-MS/MS). It was possible to identify4proteins, which comprise heat shock protein90, beta (Grp94), member1, proteindisulfide-isomerase A6, beta-actin (aa27-375), A-X actin and gamma-actin.The composition of maize pollen flavonoids was analyzed by LC-Q-TOF-MSand other analysis. The quercetol, kaempferol, isorhamnetin, quercetin 3-O-glucoside, isorhamnetin3-O-glucoside, quercetin3,3’-0-diglucoside andquercetin3,7-O-diglucoside were identified in maize pollen flavonoids’ factor.
