Momordicu grosvenori is a kind of plants of the family Cucurbitaceae, which has the function of moistening lung, eliminating phlegm, suppressing cough and has been applied to the industry of medicine. We found the optimal factor extraction condition in extract flavone from Momordicu grosvenori in the reflux extraction process by the single factor experiment、orthogonal experiments and the response surface methodology(RSM). Through the static-dynamic adsorption test and the polyamide resin to get further purification.The main results were listed as follows:1. The experiment of single factors was used to optimize the influence factors (ethanol concentration, extraction temperature, extraction time, the ratio to matial of liquid, times) in the reflux extraction process. It was found that the optimal single factor extraction condition were ethanol concentration80%, temperature80℃, extraction time1h, the ratio between material and liquid1:30, once, respectively.2. On the basis of single factor experiment, ethanol concentration, temperature, extraction time and the ration between material and liquid were chosed to do an orthogonal test. The extraction rate can reach maximum1.52%when the process was performed by1h extraction once on80℃with30-fold volume80%ethonal.3. According to the result of single factor and orthogonal experiments, we chose ethanol concentration, extraction temperature, extraction time to study the interaction between these factors through a response surface methodology (RSM). The results showed, the optimal parameters are as follows:ethanol concentration is76.59%, extraction temperature is84.29℃, extraction time is0.76h, the yield rate is1.573%.Actual production, the optimum process conditions can be adjusted as follows:ethanol concentration is77%, extraction temperature is85℃. extraction time is48min.the yield rate is1.538%.4. Through the static-dynamic adsorption test, choosing AB-8macroporous resin to determine the conditions is:the concentration of the test is2.5mg/mL, pH is4, flow rate on adsorption is2BV/h. The flow rate on desorption is2BV/h and the ethanol concentration is50%. Detection calculation to determine the purity of flavone from Mangosteen is39.7%.5. Through the polyamide resin to get further purification. Determine the optimal conditions is:the concentration of the test is30mg/mL an the flow rate is1BV/h. The flow rate on desorption is1.5BV/h an the ethanol concentration is60%. Through the HPLC analysis, the extract contains flavonoids. Detection calculation to determine the purity of flavone from Mangosteen is81.7%.
