The Casin Kinase1family(CK1)regulate multiple cellular aspects and play important roles in Wnt/Wingless and Hedgehog signaling. The kinase domain of Drosophila Gilgamesh-I, A homolog of human CK1-γ, was purified and crystallized.Crystal of Gilgamesh-I kinase domain diffracted to2.85A and belonged to space group P43212with unit-cell parameters a=b=52.025A, c=291.727A. We studied the structure of Gilgamesh-I kinase domain by replacing some molecular, and we found that the kinases domain has active conformation and conserved catalytic elements. Structural comparison indicates that an extended loop between al helix and β4strand exists in the Gilgamesh-I kinase domain.
