Tead is a highly conserved gene family during evolution. In mouse, tead4play a key function in skeletal muscle differentiation including muscle fiber fusion and a shorter muscle fiber formation. In the present study, the tead4designed ShRNA528, shRNA1097, shRNA1331and shRNA1448were inserted into the lentiviral back bone, the vectors were then transfected to bovine muscle satellite cells (bMSCs). The results showed that the tead4knockdown ratios in these cells were75.1%,36.7%,53.9%and66.4%, respectively. A robust cell line transfected by shRNA528was selected and used for further researches. In bMSCs, tead4knock-down did not affect the ability of the differentiation of cells into myotubes, osteoblasts and neurocytes. The expressions of the associated genes BGP, NESTIN and MHC did not affected when compared to the non-transfected cells. However, tead4knock-down significantly decreased the ability of trans-differentiation from muscle satellite cells to adipocytes. These results suggested that the knock-down of tead4affect the transformation of satellite cells to adipocytes.
